摘 要:目的观察心肌梗死(心梗)前、心梗后及心梗前后联合运动对大鼠心肌梗死后晚期微血管密度的影响,并检测不同运动方案对左室血管内皮细胞生长因子(VEGF)及其受体mRNA和蛋白表达的调控效应。方法采用随机数字表法将96只雄性SD大鼠分为假手术不运动组(Sed-Sh组)、假手术前运动组(PreE-Sh组)、心梗不运动组(Sed-MI组)、心梗前运动组(PreE-MI组)、心梗后运动组(PostE-MI组)及心梗前后联合运动组(ComE-MI组),每组16只。在正式实验前,各运动组大鼠先在跑台上进行适应性训练,然后给予跑台训练,每天60 min,每周训练5 d,共训练5周;而不运动组大鼠整个实验过程中均不进行运动训练。在第5周最后1次训练结束24 h后,所有大鼠均进行急性心肌梗死手术或假手术。经休息康复4周后,PostE-MI组及ComE-MI组进行5 d适应性训练,随后进行8周跑台运动,每天60 min,每周训练5 d。于实验结束时处死所有大鼠,应用Ⅷ-related Antigen染色进行左室梗死区和非梗死区毛细血管密度测定;采用实时PCR方法检测VEGF及其受体mRNA表达;采用免疫印迹法检测VEGF及其受体蛋白表达。结果与Sed-Sh组比较,PreE-Sh组微血管密度有增加趋势,但差异无统计学意义(P>0.05);与Sed-MI组比较,PostE-MI组和ComE-MI组微血管密度均明显增加(P<0.05),PreE-MI组大鼠微血管密度有增加趋势,但差异无统计学意义(P>0.05)。另外PostE-MI组微血管密度显著高于PreE-MI组(P<0.05),而ComE-MI组微血管密度显著高于PostE-MI组(P<0.05)。与Sed-MI组比较,PostE-MI组和ComE-MI组大鼠心脏VEGF及其受体mRNA表达均明显增强(P<0.05),但PostE-MI组与ComE-MI组大鼠组间差异无统计学意义(P>0.05)。与Sed-MI组比较,PostE-MI组和ComE-MI组大鼠心脏VEGF及其受体蛋白表达均明显增强(P<0.05),但PostE-MI组与ComE-MI组大鼠组间差异无统计学意义(P>0.05)。结论心梗前运动对于心梗晚期微血管密度无显著影响,而心�Objective To investigate the effect of exercise both before and after myocardial infarction(MI)on microvessel density in the left ventricle,and on the mRNA and protein expression of vascular endothelial growth factor(VEGF)and its receptors.Methods Ninety-six male Sprague-Dawley rats were randomly divided into a sedentary sham operation group(Sed-Sh),a pre-MI exercise sham operation group(PreE-Sh),a sedentary MI group(Sed-MI),a pre-MI exercise group(PreE-MI),a post-MI exercise group(PostE-MI)and a combined exercise group(ComE-MI).The rats of the exercise groups underwent 60min of treadmill training daily five times a week for five weeks,while those of the Sed-Sh and Sed-MI groups remained sedentary.Then all of the rats underwent either an operation to induce an acute MI(Sed-MI,PreE-MI,PostE-MI and ComE-MI groups)or a sham MI operation(Sed-Sh and PreE-Sh groups).Four weeks after the operation,the exercise groups underwent five days adaptive training,followed by 8 weeks of 60min of treadmill training daily,five times a week.All of the rats were then sacrificed.The microvessel densities in the infarcted and uninfarcted volumes were detected using Factor VIII-related antigen staining,while the mRNA expressions of VEGF and its receptors were examined using a real-time polymerase chain reaction.The protein expressions of VEGF and its receptors were measured using western blotting.Results Compared with Sed-MI group,the average microvessel densities of the PostE-MI and ComE-MI groups had increased significantly,with the average density of the PostE-MI group significantly higher than that of the PreE-MI group,but significantly lower than the ComE-MI group′s average.Compared with the Sed-MI group,the average mRNA expression of VEGF and its receptors in the PostE-MI and ComE-MI groups was significantly higher,without significant differences between the two groups.Conclusions Pre-MI exercise does not significantly improve left ventricular function or microvessel density,while post-MI exercise does,as it can upregulate the e
