摘  要:【目的】探究黄瓜PIP2;4(CsPIP2;4 Gene ID:101215300)在干旱胁迫响应中的可能功能。【方法】以华北型黄瓜‘9930’为试材,基于CsPIP2;4为前期研究筛选出的干旱胁迫下显著差异表达蛋白,克隆CsPIP2;4基因,对其进行生物信息学分析,并利用qRT-PCR检测CsPIP2;4在干旱下的表达。【结果】生物信息学分析显示,CsPIP2;4 CDS序列有852 bp,编码283个氨基酸,CsPIP2;4蛋白具有疏水性、无信号肽段、有跨膜结构,是定位在质膜的内在蛋白,以无规则卷曲和α-螺旋结构为主,具有典型的水通道蛋白结构功能域,属于MIP超家族;蛋白质同源比对分析显示,CsPIP2;4与甜瓜PIP2;7的亲缘关系最近。qRT-PCR结果表明CsPIP2;4表达受干旱诱导。【结论】CsPIP2;4可能是参与黄瓜干旱胁迫应答过程重要基因,为后续鉴定其在黄瓜响应干旱中的功能奠定了基础。【Objective】To explore the possible function of PIP2;4 in cucumber(CsPIP2;4 Gene ID:101215300)involvement in drought stress.【Method】Using the North China cucumber 9930 as the test material,based on CsPIP2;4 as the significantly differentially expressed protein selected under the drought stress in the previous study.The CsPIP2;4 gene was cloned and analyzed with bioinformatics,and the expression level under drought-stress was detected by Real-time Quantitative PCR.【Result】Bioinformatics analysis showed that CDS of the CsPIP2;4 was 852 bp in length,encoded a protein of 288 amino acid residues.CsPIP2;4 has hydrophobic and transmembrane domain,but it doesn't have signal peptide.CsPIP2;4 is located intrinsic protein of plasma membrane.The main secondary structure of CsPIP2;4 is random curl and α-spiral,and it has typical functional domain of aquaporins and belongs to the major intrinsic protein(MIP)family.The evolutionary analysis showed that CsPIP2;4 was the closest of Cme PIP2-7.The qPCR results showed CsPIP2;4 may be involved in the response related to drought stress.【Conclusion】CsPIP2;4 may be an important gene involved in the response process of cucumber under the drought stress,the study lays the foundation for the further function identification of CsPIP2;4 in response to drought.
