摘　　要：目的采用HPLC法同时测定银黄胶囊中8种指标性成分的含量。方法采用Waters Sunfire C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸,梯度洗脱,流速1.0 mL·min^-1,柱温30℃,检测波长分别为326 nm(新绿原酸、绿原酸、隐绿原酸、3,4-O-二咖啡酰基奎宁酸、3,5-O-二咖啡酰基奎宁酸、4,5-O-二咖啡酰基奎宁酸)、276 nm(黄芩苷、黄芩素),进样量10μL。结果新绿原酸、绿原酸、隐绿原酸、3,4-O-二咖啡酰基奎宁酸、3,5-O-二咖啡酰基奎宁酸、4,5-O-二咖啡酰基奎宁酸、黄芩苷和黄芩素分别在0.8261~8.261、4.905~49.05、0.8369~8.369、1.082~10.82、0.9407~9.407、1.209~12.09、17.12~171.2、1.361~13.61μg·mL-1范围内与峰面积呈良好的线性关系,平均回收率分别为101.27%、99.86%、98.49%、100.39%、98.48%、98.33%、98.20%、97.87%,RSD分别为1.05%、1.83%、1.79%、1.44%、0.98%、1.07%、1.31%、0.83%。结论所用方法准确、重复性好,可用于银黄胶囊中8种指标性成分的含量测定。OBJECTIVE To establish an HPLC method for simultaneous determination of eight components in Yinhuang capsules.METHODS HPLC analysis was carried out on a Waters Sunfire C18 column(250 mm×4.6 mm,5μm)with mobile phase consisting of acetonitrile-0.1%phosphoric acid at a flow rate of 1.0 mL·min^-1 in a gradient elution manner.The column temperature was 30℃and the detection wavelength was set at 326 nm for neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,3,4-O-dicaffeoylquinic,3,5-O-dicaffeoylquinic,4,5-O-dicaffeoylquinic,and 276 nm for bacicalin and baicalein.RESULTS A good linearity of neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,3,4-O-dicaffeoylquinic,3,5-O-dicaffeoylquinic,4,5-O-dicaffeoylquinic,bacicalin and baicalein was achieved in the range of 0.8261-8.261μg·mL-1,4.905-49.05μg·mL-1,0.8369-8.369μg·mL-1,1.082-10.82μg·mL-1,0.9407-9.407μg·mL-1,1.209-12.09μg·mL-1,17.12-171.2μg·mL-1,and 1.361-13.61μg·mL-1,respectively.The average recovery of eight components was 101.27%(RSD=1.05%),99.86%(RSD=1.83%),98.49%(RSD=1.79%),100.39%(RSD=1.44%),98.48%(RSD=0.98%),98.33%(RSD=1.07%),98.20%(RSD=1.31%),and 97.87%(RSD=0.83%),respectively.CONCLUSION The method is accurate and reproducible,and it can be used for the content determination of eight components in Yinhuang capsules.
