摘 要:目的:证明葫芦素B通过影响JAK2/STAT3信号活化抑制非小细胞肺癌(NSCLC)H1975细胞增殖。方法:采用不同浓度葫芦素B作用于H1975细胞不同时间,采用CCK-8法测定细胞增殖抑制情况,采用PI染色法检测细胞周期分布,采用Western blot法测定EGFR、JAK2、STAT3蛋白表达。结果:葫芦素B浓度10、100、1 000 nmol/L作用24、48 h的细胞增殖抑制率均高于葫芦素B浓度0 nmol/L,其中1 000 nmol/L>100 nmol/L>10 nmol/L(P<0.05)。葫芦素B浓度10、100、1 000 nmol/L作用48 h的细胞增殖抑制率均高于作用24 h(P<0.05)。细胞增殖抑制率随着葫芦素B浓度增加和作用时间延长逐渐增高(P<0.05)。葫芦素B浓度100、1 000 nmol/L作用24、48 h的G2/M期阻滞率均高于葫芦素B浓度0、10 nmol/L,其中1 000 nmol/L>100 nmol/L(P<0.05)。葫芦素B浓度100、1 000 nmol/L作用48 h的G2/M期阻滞率均高于作用24 h(P<0.05)。G2/M期阻滞率随着葫芦素B浓度增加和作用时间延长逐渐增高(P<0.05)。葫芦素B浓度100、1 000 nmol/L作用H1975细胞后p-STAT3与p-JAK2的表达均低于葫芦素B浓度0、10 nmol/L,其中1 000 nmol/L>100 nmol/L(P<0.05)。结论:葫芦素B能够抑制H1975细胞增殖、阻滞细胞周期,其机制与葫芦素B抑制JAK2/STAT3信号活化有关。Objective:To demonstrate that Cucurbitacin B inhibits non-small cell lung cancer (NSCLC) H1975 cell proliferation by influencing JAK2/STAT3 signaling activation.Method:H1975 cells were treated with different concentrations of Cucurbitacin B at different time periods,cell proliferation inhibition was determined by CCK-8 method,cell cycle distribution was detected by PI staining,and protein expressions of EGFR,JAK2,and STAT3 were determined by Western blot.Result:The inhibition rate of cell proliferation at Cucurbitacin B concentrations of 10,100 and 1 000 nmol/L for 24 and 48 h were higher than those at concentration of 0 nmol/L,of which 1 000 nmol/L > 100 nmol/L > 10 nmol/L (P<0.05).The inhibition rate of cell proliferation at Cucurbitacin B concentrations of 10,100 and 1 000 nmol/L for 48 h were higher than those of 24 h (P<0.05).The inhibition rate of cell proliferation increased gradually with the increase of Cucurbitacin B concentration and the prolongation of action time (P<0.05).The arrest rate of G2/M phase at Cucurbitacin B concentrations of 100 and 1 000 nmol/L for 24 and 48 h were higher than those at concentration of 0 and 10 nmol/L,in which 1 000 nmol/L > 100 nmol/L (P<0.05).The arrest rate in G2/M phase at Cucurbitacin B concentrations of 100 and 1 000 nmol/L for 48 h were higher than those at 24 h (P<0.05).The arrest rate of G2/M phase increased gradually with the increase of Cucurbitacin B concentrations and the prolongation of action time (P<0.05).The expression of p-STAT3 and p-JAK2 in H1975 cells at Cucurbitacin B concentrations of 100 and 1 000 nmol/L were lower than those in H1975 cells at concentration of 0 and 10 nmol/L,of which 1 000 nmol/L>100 nmol/L (P<0.05).Conclusion:Cucurbitacinacin B can inhibit the proliferation and cell cycle of H1975 cells,and its mechanism is related to the inhibition of JAK2/STAT3 signal activation by Cucurbitacin B.
