摘  要:目的:研究艾叶精油对小鼠肝脏Bmal1和Clock生物钟基因的影响。方法:将♀♂小鼠随机分为8组。选取早、晚不同的时间,连续7d腹腔注射给药,观察其临床表现及体重变化。实验结束,分别于10:00和晚上22:00处死小鼠,取肝脏组织实时荧光定量RT-PCR法检测Bmal1,Clock生物钟基因的水平。结果:艾叶油可以促进小鼠Bmal1,Clock生物钟基因的表达,且艾叶油在10:00给药时比22:00给药效果更明显。此外雄鼠比雌鼠对艾叶油更敏感。结论:艾叶油早晚给药对生物钟基因影响具有显著不同,且具有性别差异,可为其临床应用提供理论参考。Objective To study the effect of Artemisiae argyi oil on the expression of Bmal1 and Clock in the liver of mice.Methods Mice were randomly divided into eight groups,half male and half female.Each group were respectively injected with Artemisiae argyi oil at 10∶00 a.m.and 10∶00 p.m.for seven days.Clinical manifestations and weight changes were observed and recorded.After the experiment,the mice were respectively killed at 10∶00 a.m.and 10∶00 p.m.Real time-PCR was used to detect the expression of Bmal1 and Clock in the liver of mice.Results Artemisiae argyi oil promotes the expression of Bmal1 and Clock in mice,and Artemisiae argyi oil was more effective when administered at 10∶00 a.m.than 10∶00 p.m.,in addition,males were more sensitive to Artemisiae argyi oil than females.Conclusion Artemisiae argyi oil administered in the morning and evening has significantly different effects on circadian rhythm and genders of mice,which can provide a theoretical reference for its clinical application.
