摘  要:【目的】本文为了解AraC家族转录调控因子Lmo2672对单核增生李斯特菌(LM)环境适应性和应激的影响。【方法】利用温控型质粒pKSV7构建LM-Δlmo2672缺失株,比较母源株LM EGD-e和缺失株LM-Δlmo2672在不同应激环境中生长情况的差异,并利用qRT-PCR方法分析转录调控因子Lmo2672缺失对应激调控基因及氧化应激相关基因转录水平的影响。【结果】与LM EGD-e相比,LM-Δlmo2672在37、42℃及含5%NaCl、0.3%胆酸盐、5 mM H2O2、1%Triton X-100的培养基中生长速度均显著降低(P<0.05);且环境适应性及应激相关基因(prfA、lexA、fri、perR、kat、sod)转录水平显著降低(P<0.05)。【结论】提示lmo2672基因在LM环境适应性和应激中发挥重要的调控作用。【Objective】The present paper aimed to determine the effect of the AraC family transcriptional regulator lmo2672 on the environmental adaptability and stress.【Method】AΔlmo2672 mutant was constructed using temperature-sensitive shuttle plasmid pKSV7,and the environmental adaptability of LM-Δlmo2672 with wild-type strain LM EGD-e,was also compared.Then,the effects of lmo2672 deletion on the transcriptional levels of genes related to stress regulator and oxidative stress of LM using qRT-PCR were explored.【Result】Compared with LM EGD-e,LM-Δlmo2672 grew significantly slowly at 37,42℃,and media containing 5%NaCl,0.3%bile salts,5 mM H2O2,1%Triton X-100(P<0.05).Moreover,the transcription levels of regulatory genes and oxidative stress-related genes(prfA,lexA,fri,perR,kat,sod)were significantly decreased(P<0.05).【Conclusion】These findings indicated that lmo2672 played important regulatory roles in the environmental adaptability and stress.
