摘 要:目的观察骨髓间充质干细胞(mesenchymal stem cells,MSCs)联合肥胖抑制素(obestatin,OB)观察其对Ⅱ型糖尿病(type 2 diabetes mellitus,T2DM)小鼠实验模型的影响以及对T2DM小鼠胰腺β细胞内分泌功能损伤的治疗潜力。方法制备T2DM小鼠模型。另取正常小鼠,分离骨髓MSCs并进行细胞培养并移植。测定各组动物的空腹血糖、空腹血清胰岛素和血清C肽水平。检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)。计算胰岛素抵抗指数(HOMA-IR)和胰岛β细胞功能指数(HOMA-β)。免疫组织化学方法检测CD105,Bax和胰岛素在胰腺组织的表达情况。结果T2DM小鼠HOMA-IR,血清葡萄糖和血脂水平显著增加,HOMA-β、胰岛素水平显著的下降。MSCs或OB单独治疗在体重、体重指数、血清葡萄糖、TC、TG、LDL、HOMA-IR和HOMA-β等方面对T2DM小鼠都有保护作用,体现在以上所有参数上均有显著改善。MSCs和OB联合应用治疗效果更显著。MSCs和OB的治疗组Bax表达较T2DM组动物降低,同时改善了动物的代谢水平。结论移植MSCs能部分重建胰岛组织,恢复胰岛功能,有效改善T2DM实验模型的高血糖和高脂血症,联合OB能达到更好的治疗效果。Objective To observe the effect of bone marrow mesenchymal stem cells(MSCs)combined with bestatin(OB)on the experimental mouse models with type 2 diabetes mellitus(T2DM)and its therapeutic potential for endocrine function injury of beta cells in T2DM mice.Methods The experimental mouse models with T2DM were prepared.Normal mice were selected,whose bone marrow MSCs were isolated,cultured,and transplanted.The levels of fasting blood glucose,fasting serum insulin and serum C-peptide in each group were determined.Serum total cholesterol(TC),triglycerides(TG),high-density lipoprotein cholesterol(HDL-c)and low-density lipoprotein cholesterol(LDL-c)were measured.HOMA-IR and HOMA-βwere calculated.The expression of CD105,Bax and insulin in pancreatic tissue was detected by immunohistochemistry.Results HOMA-IR,serum glucose and lipid levels significantly increased,while HOMA-βand insulin levels significantly decreased in T2DM mice.The single use of MSCs or obestatin had a protective effect on diabetic mice in body weight,body mass index,serum glucose,TC,TG,LDL,HOMA-IR and HOMA-β,with significant improvement in all the above parameters.The combination of MSCs and obestatin was more effective.The expression of Bax in the treatment group of MSCs and OB was lower than that in the T2DM group,and the metabolic level of the animals was improved.Conclusion The transplantation of MSCs can partially reconstruct islet tissue,restore islet function,effectively improve hyperglycemia and hyperlipidemia in the experimental models with T2DM,and achieve better therapeutic effect in combination with obestatin.
