摘  要:目的:建立测定人血浆中米卡芬净浓度的高效液相色谱法。方法:以氯唑沙宗为内标,血浆样品用乙腈沉淀蛋白处理后,经C18色谱柱进行分离,流动相为乙腈-5 mmol/L醋酸铵(V∶V=40∶60),流速为1.0 ml/min,柱温为35℃,检测波长为273 nm,同时进行方法学的验证,并将其应用于重症患者血浆中米卡芬净浓度的测定。结果:米卡芬净在0.2~25.0μg/ml浓度范围内线性关系良好,最低定量下限为0.2μg/ml,低、中及高浓度质量控制样品的回收率分别为88.54%、83.27%及88.16%,日内和日间精密度均在±15%之内。结论:本方法简单快速、准确可靠,可用于临床重症患者米卡芬净血药浓度的监测。OBJECTIVE:To establish a HPLC method for determining the concentration of micafungin in human plasma.METHODS:Chlorzoxazone was set as internal standard substance,the plasma sample was proceeded by acetonitrile protein precipitation and isolated by C18 chromatographic column,with the mobile phase was acetonitrile-5 mmol/L of ammonium acetate(V∶V=40∶60),the flow rate was 1.0 ml/min,the column temperature was 35℃and the detecting wavelength was 273 nm,methodological verification was conducted and applied to determine the concentration of micafungin in human plasma in critical patients.RESULTS:Micafungin was in a good linear relationship in the concentration range of 0.2-25.0μg/ml,with the lowest quantification limit was 0.2μg/ml.The recovery rates of low,medium and high concentration quality control samples were respectively 88.54%,83.27%and 88.16%,the intra-day and inter-day precision were both within±15%.CONCLUSIONS:This method is simple,fast,accurate and reliable,which can be used to monitor the blood concentration of micafungin in critical patients.
