摘 要:为探讨共激活相关精氨酸甲基转移酶1(coactivator-associated arginine methyltransferase 1,CARM1)对食管癌细胞增殖和迁移能力的影响,采用免疫组织化学方法检测食管癌组织和癌旁组织中CARM1的表达情况,利用特异的短发夹RNA(short hairpin RNA,shRNA)分别敲低食管癌细胞株KYSE510和TE-1中CARM1的表达,通过过表达质粒上调食管癌细胞株KYSE150和ECA9706中CARM1的表达,采用CCK-8生长曲线试验和平板克隆形成试验检测CARM1敲低和上调后的食管癌细胞增殖能力的变化情况,并采用Transwell细胞迁移试验检测细胞迁移能力的变化情况。结果显示,与癌旁组织比较,CARM1蛋白在食管癌组织中高表达(P<0.01);敲低CARM1后食管癌细胞的迁移和增殖能力减弱(P<0.05);过表达CARM1后食管癌细胞的迁移和增殖能力增强(P<0.05)。提示CARM1在促进食管癌细胞生长和恶性肿瘤进展过程中发挥重要作用。Our purpose is to explore the effects of coactivator-associated arginine methyltransferase 1(CARM1) on the proliferation and migration of esophageal cancer cells. Firstly, the expression of CARM1 in esophageal cancer and normal tissues was detected by immunohistochemical staining. Then CARM1 was knocked down by short hairpin RNA(shRNA) or overexpressed by overexpressed plasmids in different esophageal cancer cell lines, respectively. The proliferation of esophageal cancer cells was evaluated by CCK-8 assay and colony formation assay. The migration ability of esophageal cancer cells was measured by Transwell chamber. The results showed that, compared with adjacent tissues, CARM1 was highly expressed in esophageal cancer tissues(P<0.01). The proliferation and migration ability of esophageal cancer cells were significantly inhibited when CARM1 was knocked down by shRNA(P<0.05). The proliferation and migration ability of esophageal cancer cells were significantly enhanced when CARM1 was overexpressed(P<0.05). We conclude that CARM1 may play an important role in promoting cell proliferation and the development of esophageal cancer.
