摘 要:目的:研究复方当归注射液对对叔丁基过氧化氢(t-BHP)诱导人脐静脉内皮细胞(EA.hy926)氧化损伤的保护作用。方法:首先测定DPPH自由基及羟基自由基清除率,初步研究复方当归注射液的体外抗氧化能力;然后体外培养EA.hy926细胞,分为正常组、模型组(t-BHP组)和复方当归注射液处理组(各浓度复方当归注射液+t-BHP组),采用MTT法检测各组细胞活力,试剂盒比色法检测各组乳酸脱氢酶(LDH)、丙二醛(MDA)、超氧化物歧化酶(SOD)、还原性谷胱甘肽(GSH-Px)含量。结果:复方当归注射液清除DPPH及羟基自由基有显著的清除效果,EC50值分别为20.88μL/mL,0.8211μL/mL。对比正常组,t-BHP(600μmol/L)可显著致EA.hy926细胞氧化损伤,经复方当归注射液预处理可显著提高EA.hy926细胞的活性,降低LDH释放量和MDA含量,提高SOD和GSH-Px活力,并呈明确的浓度依赖性。结论:复方当归注射液对t-BHP诱导的EA.hy926细胞氧化损伤具有保护作用。Objective:To investigate the protective effect of Compound Angelica Injection(CAI)against tert-butyl hydroperoxide(t-BHP)-induced Human umbilical vein endothelial cells(EA.hy926)injury.Methods:Firstly,the antioxidant activity of compound angelica injection was evaluated by scavenging capabilities of DPPH·and·OH free radicals.Then EA.hy926 cells were randomly divided into control group,injured group(stimulated by t-BHP),and CAI treatment group(pretreated with various dose of CAI followed by t-BHP stimulation).The viability of each group was detected by MTT assay.The levels of lactate dehydrogenase(LDH),malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were measured by colorimetry kit.Results:CAI had obvious effect on scavenging ability of DPPH·and·OH free radicals,with EC50 values of 20.88μL/mL and 0.8211μmL/mL,respectively.Compared with the control group,t-BHP(600μmol/L)caused significantly oxidative damage in EA.hy926 cells.Compared with t-BHP group,the survival rate of the CAI treatment group gradually increased in a dose-dependent manner.In addition,CAI inhibited the increase of LDH and MDA induced by t-BHP and increased the activity of SOD and GSH-Px in a concentration-dependent manner.Conclusion:CAI has protective effects on t-BHP-induced oxidative damage in EA.hy926 cells.
