摘  要:目的:观察电针神庭、百会穴对大脑中动脉闭塞模型(MCAO)大鼠神经功能及海马中CACNα1D mRNA表达的影响,探讨其可能的作用机制。方法:将SD大鼠随机分为电针组、模型组、假手术组。采用Longa改良线栓法制作MCAO大鼠模型,造模后24 h后电针组每天电针神庭、百会穴20 min,连续7 d,模型组、假手术组予同等条件抓取。通过神经功能缺损评分判断大鼠神经功能缺损状况;2,3,5-氯化三苯基四氮唑(TTC)染色观察脑梗死面积;实时定量荧光PCR(q-PCR)检测脑组织CACNα1D mRNA表达。结果:电针组神经功能缺损评分高于假手术组,低于模型组(P<0.05);脑梗死面积观察可见电针组脑部灰白色梗死灶大于假手术组,小于模型组;电针组海马中CACNα1D mRNA表达高于假手术组,低于模型组(P<0.05)。结论:电针能改善MCAO大鼠的神经功能状况,其机制可能与下调CACNα1D mRNA在海马中的表达有关。Objective:To observe the effect of electroacupuncture at Shenting and Baihui points on nerve function and the expression of CaCNα1D mRNA in hippocampus of MCAO rats,and to explore its possible mechanism.Methods:SD rats were randomly divided into electroacupuncture group,model group and sham operation group.MCAO rat model was constructed by Longa modified thread bolting method.After 24 hours of modeling,the electroacupuncture group received electroacupuncture in Shenting and Baihui acupoints for 20 minutes every day for 7 consecutive days.The model group and the sham operation group were captured under the same conditions.The nerve function defect of rats was judged by the nerve function defect score.Cerebral infarction area was observed by staining with 2,3,5-triphenyltetrazolium chloride(TTC).Real-time quantitative fluorescent PCR(q-PCR)was used to detect the expression of CACNα1D mRNA in brain tissue.Results:The neurological deficit score of EA group was higher than that of sham operation group and lower than that of model group(P<0.05);the gray infarct area of EA group was larger than that of sham operation group and smaller than that of model group;the expression of CaCNα1D mRNA in hippocampus of EA group was higher than that of sham operation group and lower than that of model group(P<0.05).Conclusion:Electroacupuncture can improve the neurological function of MCAO rats,and the mechanism may be related to the down-regulation of the expression of CACNα1D mRNA in the hippocampus.
