摘 要:目的:测定山西不同产地野生款冬花中款冬酮的含量。方法:样品以乙醇回流提取后,采用高相液相色谱法测定款冬酮的含量。色谱柱为Alltima C18(250 mm×4.6 mm,5μm);流动相采用甲醇-水溶液(85∶15)等度洗脱,流速为1.0 mL/min,柱温为30℃;检测波长220 nm。结果:款冬酮进样量在0.52~6.24μg范围内呈良好线性关系,回归方程y=2631856.73x-4617.38,r=0.9993,平均回收率为98.03%,RSD为1.56%。结论:不同产地野生款冬花中款冬酮的含量不同,为款冬花药材的规范化栽培提供科学依据。Objective:To determine the contents of tussilagone in wild Farfarae Flos from different habitats in Shanxi province.Methods:After reflow extraction by ethanol,the contents of tussilagone in samples were determined by HPLC.The determination was performed on Alltima C18(250 mm×4.6 mm,5μm)column with mobile phase consisted of methanol-water(85∶15)at the flow rate of 1.0 mL/min.The column temperature was 30℃and the detection wavelength was 220 nm.Results:The linear relations of tussilagone was good in the range of 0.52~6.24μg.The regression equation was y=2631856.73x-4617.38,r=0.9993.The average recovery was 98.03%and RSD was 1.56%.Conclusion:The content of tussilagone is different in samples from different habitats.The results can provide scientific basis for standard cultivation of Pueraria lobata.
