摘　　要：目的:研究金刚藤对人肝癌SMMC-7721细胞自噬相关蛋白LC3的影响。方法:体外培养人肝癌SMMC-7721细胞,分为空白对照组及金刚藤低、中、高剂量组,分别给予空白血清、金刚藤含药血清(0.5 g/ml、1 g/ml、2 g/ml)培养48 h后,采用CCK-8法测定各组SMMC-7721细胞增殖的抑制率;Western Blot法检测各组LC3-Ⅰ、LC3-Ⅱ蛋白相对表达量的表达。结果:与空白对照组比较,金刚藤低、中、高剂量组细胞增殖抑制率均显著增加(均P<0.05),金刚藤各剂量组随着浓度增高而抑制率增高(均P<0.05),呈浓度依赖性;与空白对照组比较,金刚藤各剂量组LC3-Ⅰ蛋白相对表达量无显著性差异,LC3-Ⅱ蛋白相对表达量均显著增加(均P<0.05),LC3-Ⅱ/LC3-Ⅰ比值均显著增高(均P<0.05)。结论:金刚藤可抑制人肝癌SMMC-7721细胞增殖,可能与其调控自噬有关。Objective:To study the effect of Smilax China L.on the autophagy-related protein LC3 of Human hepatocellular carcinoma SMMC-7721 cells.Methods:Human hepatocellular carcinoma SMMC-7721 cells were cul⁃tured in vitro and divided into blank control group,low,medium and high dose groups of Smilax China L.,blank se⁃rum and Smilax China L.serum(0.5 g/ml,1 g/ml,2 g/ml)After 48 hours of culture,the inhibition rate of SMMC-7721 cell proliferation in each group was determined by CCK-8 method;the relative expression of LC3-Ⅰ and LC3-Ⅱ protein in each group was detected by Western blot method.Results:Compared with the blank control group,the inhi⁃bition rate of cell proliferation in the low,medium and high groups of Smilax China L.increased significantly(P<0.05,all groups),and the inhibition rate of each group of Smilax China L.increased as well as concentration(P<0.05,all groups),showing a concentration dependence.Compared with the blank control group,the relative ex⁃pression of LC3-Ⅰprotein in each dose group of Smilax China L.was not significantly different,and the relative expres⁃sion of LC3-Ⅱprotein in the low,middle and high groups of Smilax China L.increased significantly(P<0.05,all groups),LC3-Ⅱ/LC3-Ⅰratios all increased significantly(P<0.05,all groups).Conclusion:Smilax China L.can in⁃hibit the proliferation of Human hepatocellular carcinoma SMMC-7721 cells,the mechanism may be related to the regulation of autophagy.
