摘 要:目的:探讨扶阳活血解毒方对溃疡性结肠炎(UC)模型大鼠的治疗作用及血清白细胞介素-17(IL-17)、白细胞介素-10(IL-10)水平的影响。方法:将60只大鼠随机分为对照组、模型组、柳氮磺吡啶组、扶阳活血解毒方高剂量组、扶阳活血解毒方中剂量组、扶阳活血解毒方低剂量组,每组10只。采用三硝基苯磺酸(TNBS)/乙醇法造模,治疗14 d后,观察大鼠一般状况并进行疾病活动指数(DAI)评分,酶联免疫吸附(ELISA)法检测大鼠血清IL-17、IL-10水平变化,取结肠标本观察结肠组织病理学变化。结果:与对照组比较,模型组大鼠DAI评分以及血清IL-17水平显著增加,血清IL-10水平明显降低,结肠组织HE染色病理切片提示炎症细胞浸润,结肠组织黏膜层、黏膜下层均有不同程度的损伤;与模型组比较,扶阳活血解毒方高、中、低剂量组大鼠DAI评分以及血清IL-17水平明显降低,血清IL-10水平明显增加,差异具有统计学意义(P<0.01);结肠组织病理切片示扶阳活血解毒方各组炎症细胞浸润减少或消失,结肠组织黏膜层、黏膜下层的损伤均得到不同程度的改善。结论:扶阳活血解毒方对UC模型大鼠症状及结肠组织损伤有明显的改善作用,可能与降低血清IL-17水平,升高血清IL-10水平有关。Objective:To investigate the therapeutic effect of Fuyang Huoxue Jiedu Formula(扶阳活血解毒方)on rat models with ulcerative colitis(UC)and the effects on serum levels of interleukin-17(IL-17)and interleukin-10(IL-10).Methods:Trinitrobenzene sulfonic acid(TNBS)/ethanol method was used to establish rat models with UC.Sixty rats were randomly divided into normal control group,model group,sulfasalazine group,Fuyang Huoxue Jiedu Formula high dose group,Fuyang Huoxue Jiedu Formula medium dose group,Fuyang Huoxue Jiedu Formula low dose group,10 rats in each group.After 14 days of treatment,the general condition of the rats were observed and scores of disease activity index(DAI)were evaluated.Enzyme linked immunosorbent assay(ELISA)was applied to detect the changes of serum levels of IL-17 and IL-10.Colon tissues were sampled to observe the pathological changes of the colon.Results:Compared with the normal control group,in the model group DAI scores and serum levels of IL-17 were significantly increased,serum levels of IL-10 were significantly reduced,and hematoxylin-eosin(HE)staining of colon tissues showed inflammatory cells infiltration,the damages in mucosal layer and submucosa of the colon tissue with varied degrees.Compared with the model group,the DAI score and serum levels of IL-17 in Fuyang Huoxue Jiedu formula high dose group,medium dose group and low dose group were significantly reduced,and the serum levels of IL-10 was significantly increased,with statistical difference(P<0.05).Pathological examinations of colon tissue showed a decrease or disappearance of inflammatory cell infiltration,and reduced damages to the mucosal layer and submucosa of the colon tissue with varied degrees.Conclusion:Fuyang Huoxue Jiedu Decoction can obviously improve the symptoms and histopathological examinations of rat models with UC.Reducing serum levels of IL-17 and increasing serum levels of IL-10.
