摘 要:目的探讨醛糖还原酶(aldose reductase, AR)和晚期糖化终产物受体(receptor foradvanced glycation end products, RAGE)在大鼠糖尿病视网膜病变神经细胞凋亡中的作用及其机制。方法 Wistar大鼠40只随机分为对照组、模型组、AR转染组和RAGE转染组,每组10只。模型组、AR转染组和RAGE转染组采用腹腔注射链脲佐菌素5 mL构建糖尿病大鼠模型,对照组腹腔注射等量生理盐水。模型建立1周后,AR转染组大鼠转染AR真核过表达质粒,RAGE转染组转染Rage过表达质粒pLenti-C-mGFP Rage,模型组和对照组不进行转染。检测大鼠空腹血糖,采用荧光分光光度计测定大鼠视网膜神经元细胞AR活性,采用反转录PCR法检测视网膜神经元细胞Rage mRNA相对表达量,采用原位凋亡法(TUNEL)检测视网膜神经元细胞凋亡情况,采用Western blot法检测视网膜神经元细胞Bcl-2、Bax、caspase-3、caspase-9蛋白相对表达量。结果模型组、AR转染组、RAGE转染组大鼠体质量和视网膜神经元细胞Bcl-2蛋白相对表达量低于对照组(P<0.05),空腹血糖水平及视网膜神经元细胞Bax、caspase-3、caspase-9蛋白相对表达量高于对照组(P<0.05);AR转染组和RAGE转染组大鼠体质量和视网膜神经元细胞Bcl-2蛋白相对表达量低于模型组(P<0.05),空腹血糖水平及视网膜神经元细胞Bax、caspase-3、caspase-9蛋白相对表达量高于模型组(P<0.05);AR转染组与RAGE转染组比较差异无统计学意义(P>0.05)。模型组、AR转染组、RAGE转染组大鼠视网膜神经元细胞AR活性[(3.79±0.26)、(4.21±0.48)、(3.67±0.31)u]高于对照组[(1.23±0.09)u](P<0.05),AR转染组高于RAGE转染组和模型组(P<0.05),RAGE转染组与模型组比较差异无统计学意义(P>0.05)。模型组、AR转染组、RAGE转染组大鼠视网膜神经元细胞Rage mRNA相对表达量(3.62±0.06、3.53±0.05、4.73±0.05)高于对照组(1.08±0.04)(P<0.05),RAGE转染组高于AR转染组和模型组(P<0.05),AR转染组与模型�Objective To investigate the roles of aldose reductase(AR) and receptor for advanced glycation end products(RAGE) in the neuronal apoptosis in rats with diabetic retinopathy and the mechanism. Methods Forty Wistar rats were randomly divided into control group, model group, AR transfection group and RAGE transfection group, with 10 rats in each group. Diabetic rat models were established by intraperitoneal injection of 5 mL streptozotocin in model group, AR transfection group and RAGE transfection group. Control group was intraperitoneally injected with equivalent volume of normal saline. One week after modeling, AR transfection group was transfected with AR eukaryotic overexpression plasmid, RAGE transfection group was transfected with Rage overexpression plasmid pLenti-C-MGFP Rage, and model group and control group were not transfected with plasmid. The level of fasting plasma glucose was measured, the AR activity of retinal neurons was measured by fluorescence spectrophotometer, the relative expression of Rage mRNA was detected by reverse transcription PCR, the neuronal apoptosis was detected by TUNEL, and the relative expressions of Bcl-2, Bax, caspase-3 and caspase-9 proteins in retinal neurons were detected by Western blot. Results The body mass and the relative expression of Bcl-2 protein were lower, and the fasting plasma glucose level and the relative expressions of Bax,caspase-3 and caspase-9 proteins were higher in model group,AR transfection group and RAGE transfection group than those in control group(P<0.05);the body mass and the relative expression of Bcl-2 protein were lower while the fasting plasma glucose level and the relative expressions of Bax,caspase-3 and caspase-9 proteins were higher in AR transfection group and RAGE transfection group than those in model group(P<0.05);there were no significant differences in the above indexes between AR transfection group and RAGE transfection group(P>0.05).The AR activity was higher in model group((3.79±0.26)u),AR transfection group((4.21±0.48)u)and RA
