摘 要:目的探讨前列腺素(prostaglandin, PG)E2受体和PGF2α受体在人表皮角质形成细胞上的表达及PG对人角质形成细胞病理生理的调控机制。方法 5例包皮环切术患者,取其正常包皮组织,采用免疫荧光双标记染色检测人包皮组织中表皮角质形成细胞胞膜和胞质中PGE2受体EP1、EP2、EP3、EP4蛋白及PGF2α受体FP蛋白表达情况,采用激光扫描共聚焦显微镜技术观察人包皮组织角质形成细胞中PGE2与PGF2α受体的定位情况。结果人包皮组织中表皮角质形成细胞胞膜和胞质中EP1、EP2、EP3、EP4及FP受体蛋白均阳性表达;PGE2受体表达以棘细胞层和颗粒层明显,PGF2α受体表达以棘细胞层为主。结论 PG可通过EP1、EP2、EP3、EP4及FP受体蛋白转导通路调控角质形成细胞功能。Objective To investigate the expressions of prostaglandin(PG) E2(PGE2) receptor and PGF2α receptor in human epidermal keratinocytes, and the regulation mechanism of PG in the pathophysiology of human epidermal keratinocytes. Methods The normal prepuce tissue of 5 patients undergoing circumcision was taken and detected the expressions of PGE2 receptor EP1, EP2, EP3 and EP4 proteins as well as PGF2α receptor FP protein in cytomembrane and cytoplasm of keratinocytes in human foreskin tissue by double immunofluorescence staining technique. The locations of PGE2 and PGF2α receptors in keratinocytes of human foreskin tissue were observed by laser scanning confocal microscope. Results EP1, EP2, EP3, EP4 and FP proteins were positively expressed in the cytomembrane and cytoplasm of keratinocytes in human foreskin tissue. The expression of PGE2 receptor was obvious in the stratum spinosum and the stratum granulosum, and the expression of PGF2α receptor was mainly located in the stratum spinosum. Conclusion PG modulates the function of human epidermal keratinocytes by EP1, EP2, EP3, EP4 and FP proteins pathways.
