正交试验优选紫白栓中大黄的提取工艺

摘　　要：目的优选紫白栓中大黄的提取工艺。方法以盐酸总蒽醌的含量为评价指标,采用正交试验L9(34)设计考察药材粒度、乙醇浓度、乙醇用量,筛选得出最佳提取工艺,含量测定方法用高效液相色谱法。结果以总蒽醌量计,大黄蒽醌最佳提取工艺为过20目筛大黄粉末,以70%乙醇10倍药量,浸泡30min,75℃回流30min,提取次数为3次。结论该提取工艺简单合理、稳定、切实可行。OBJECTIVE To optimize the extraction process of rhubarbs from ZiBai bolt.METHODS With the total anthraquinones as content evaluation indicator,orthogonal experiment design was used to study the Herbs particle size,the ethanol concentration,the amount of ethanol,to select out the best extraction process.The content was determined by HPLC method.RESULTS The best extraction technology of anthraquinone from Rheum palmatum L.was 20 mesh sieve powder of Rheum palmatum L.according to the total anthraquinone content,with 70%ethanol 10 times the amount of medicine,soak for 30 minutes,75℃reflux 30 min,extraction times for 3 times.CONCLUSION The extraction process is simple,stable,reasonable and feasible.
