摘  要:目的建立高效液相色谱(HPLC)快速测定人血浆中万古霉素血药浓度的方法。方法血浆样品经沉淀后,以甲醇-水(含0.05mol·L^-1磷酸二氢钾,pH=3.05)=20∶80为流动相,流速为1.0mL·min^-1,色谱柱为Agilent 5HC-C18(250×4.6mm,5μm),柱温为30℃,紫外检测波长为236nm。结果该法专属性较好,血清中杂质不干扰样品峰的测定;万古霉素在1.0~60.0mg·L^-1质量浓度范围内线性关系良好,所获回归方程为Y=17.68X+5.54(R^2=0.9999);批内、批间精密度和准确度均小于10%。结论HPLC灵敏、快速、准确,操作简便、线性范围宽,可用于万古霉素的临床血药浓度监测。OBJECTIVE To establish a method for determining the concentration of vancomycin in humanplasma by HPLC.METHODS Plasma samples were precipitated with methanol and separation was carried out on a Agilent 5 HC-C18(250×4.6 mm,5μm)at 30℃.The mobile phase consisted of methanol-water(0.05 mol·L^-1 potassium dihydrogen phosphate,pH=3.05)(20∶80)at a flow rate of 1.0 mL·min^-1.The detecting wave length was 236 nm.RESULTS The intrinsic impurity of serum did not interfere with the determination of the analyte.The linearity was good in the concentration range of 1.0-60.0 mg·L^-1 and the linear relationship was Y=17.68 X+5.54(R^2=0.9999).The precision and accuracy within batches and between batches were both less than 10%.CONCLUSION The established HPLC method is fast,sensitive,and accurate,which can be applied to the plasma concentration monitoring of vancomycin.
