高效液相色谱法测定痔复康颗粒中芦丁的含量

摘　　要：目的建立测定痔复康颗粒中芦丁含量的高效液相色谱方法。方法色谱柱采用InertSustain AQ-C18(4.6mm×250mm,5μm);乙腈-0.1%甲酸水溶液为流动相,梯度洗脱;流速:1.0mL·min^-1;检测波长:254nm;柱温:35℃;进样量:10μL。结果芦丁进样量在2.5~160μg·mL^-1范围内与峰面积呈现良好的线性关系(r=0.9999);平均加样回收率为99.78%(RSD为1.38%,n=6);4批供试品中芦丁平均含量为0.28625mg·g^-1。结论该测定方法操作简便、准确,可用于痔复康颗粒中芦丁的含量分析。OBJECTIVE Our study aims to establish a HPLC method for determination of Rutin in ZhiFuKang Granules.METHODS The InertSustain AQ-C18 column(4.6 mm×250 mm,5μm)was used.Acetonitrile and 0.1%methanoic acid solution were eluted by gradient elution.The flow rate was 1.0 mL·min^-1,the detection wavelength was set at 254 nm,the column temperature was 35℃and the injection volume was 10μL.RESULTS The responses of Rutin was linear in the range of 2.5-160μg·mL^-1(r=0.9999).The average recovery of Rutin was 99.78%(RSD was 1.38%,n=6).The average result of Rutin content in 4 samples was 0.28625 mg·g^-1.CONCLUSION The established method is simple and accurate,which can be used for the content determination of Rutin in Zhifukang Granulas.
