妇科千金片对慢性盆腔炎大鼠炎性因子及血管内皮功能相关因子的影响

摘　　要：目的探讨妇科千金片对慢性盆腔炎大鼠炎性因子及血管内皮功能相关因子的影响。方法将60只SD大鼠随机分为6组[正常对照组、假手术组、模型对照组、阳性药对照组(妇炎康,0.41 g/kg)及妇科千金片高、低剂量组(2.08,1.04 g/kg)],每组10只。各组大鼠分别灌胃相应药物,正常对照组、假手术组和模型对照组大鼠灌胃等体积生理盐水。1次/日,连续4周。末次给药后,采集各组大鼠腹主动脉血液,检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素6(IL-6)、C反应蛋白(CRP)等炎性因子水平;同时摘取子宫组织,采用Western blotting法和Real-time PCR法检测血清中炎性因子TNF-α、IL-6、CRP及子宫组织中血管内皮功能相关因子细胞间黏附分子1(ICAM-1)、血管细胞间黏附分子1(VCAM-1)、基质金属蛋白酶2(MMP-2)蛋白及基因表达水平。结果给药4周后,与假手术组比较,模型对照组大鼠血清中TNF-α,IL-6,CRP水平均显著升高,子宫组织中VCAM-1,ICAM-1,MMP-2蛋白及基因表达量均显著升高(P<0.05);与正常对照组比较,假手术组上述指标组间差异均不显著(P>0.05);与模型对照组比较,阳性对照组、妇科千金片高、低剂量组大鼠血清中TNF-α,IL-6,CRP水平均显著降低,子宫组织中VCAM-1,ICAM-1,MMP-2蛋白及基因表达量均显著降低(P<0.05);妇科千金片高、低剂量组与阳性对照组比较,上述指标差异均不显著(P>0.05)。结论妇科千金片对盆腔炎大鼠的病理状态有改善作用,作用机制可能与调控血清炎性因子及子宫组织血管内皮功能相关因子有关。Objective To investigate the effect of Fuke Qianjin Tablets on the inflammatory factors and vascular endothelial function factors in rats with chronic pelvic inflammatory disease.Methods Totally 60 SD rats were randomly divided into 6 groups:normal control group,sham-operation group,model control group,positive drug control group(Fuyankang,0.41 g/kg),Fuke Qianjin Tablets high-dose group(2.08 g/kg)and low-dose group(1.04 g/kg),10 rats in each group were given corresponding drugs by gastric perfusion respectively.The rats in the normal control group,sham-operated group and model group were given equal volume of normal saline by gavage,once a day for 4 consecutive weeks.After the last administration,abdominal aorta blood was collected to detect the levels of inflammatory factors such as tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),C-reactive protein(CRP)in serum.The uterus was extracted,and the the levels of inflammatory factors such as TNF-α,IL-6,CRP in serum and the expression levels of vascular endothelial function related factors such as intercellular adhesion molecule-1(ICAM-1),vascular ceu adhesion molecule-1(VCAM-1),matrix metalloproteinase-2(MMP-2)protein and gene in uterus tissue were detected by Western blotting and Real-time PCR.Results After 4 weeks of administration,compared with the sham-operated group,the levels of TNF-α,IL-6 and CRP in serum of the model group were significantly increased,and the expression levels of VCAM-1,ICAM-1 and MMP-2 protein and gene in uterus tissue were significantly increased(P<0.05),but the above indexes between the sham-operated group and normal control group had no significant difference(P>0.05).Compared with the model control group,the levels of TNF-α,IL-6 and CRP in serum of the positive control group,the high-and low-dose groups of Fuke Qianjin Tablets were significantly decreased,and the expression levels of VCAM-1,ICAM-1 and MMP-2 protein and gene in uterine tissue were significantly decreased,but the above indexes among the the high-and low-dose groups o
