摘 要:目的建立同时测定益肾壮骨丸中芍药苷、莫诺苷和马钱苷含量的高效液相色谱法。方法色谱柱采用Eclipse XDB-C18柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-0.3%磷酸水溶液(B)(梯度洗脱,0~20 min时7%A,20~50 min时7%A→20%A,50~55 min时20%A,55~60 min时20%A→7%A,60~70 min时7%A),流速为1 m L/min,芍药苷检测波长为230 nm、莫诺苷和马钱苷检测波长为240 nm,柱温为30℃,进样量为10μL。结果芍药苷、莫诺苷和马钱苷分别在29.30~586.00μg/m L(r=0.9998),2.94~58.74μg/m L(r=0.9997),15.29~305.70μg/m L(r=1.0000)范围内与峰面积线性关系良好;平均加样回收率分别为97.93%,99.26%,101.45%,RSD分别为0.93%,0.60%,1.40%(n=6);3批次样品中芍药苷、莫诺苷和马钱苷的含量范围分别为1.759~1.904 mg/g,0.169~0.188 mg/g,0.512~0.527 mg/g。结论该方法灵敏度高,专属性强,可用于益肾壮骨丸中芍药苷、莫诺苷、马钱苷的含量测定。Objective To establish a high performance liquid chromatography(HPLC)method for the content determination of paeoniflorin,morroniside and loganin in Yishen Zhuanggu Pills.Methods Eclipse XDB-C18 column(250 mm×4.6 mm,5μm)was used,the mobile phase was acetonitrile(A)-0.3%phosphoric acid aqueous solution(B)with gradient elution(0-20 min,7%A;20-50 min,7%→20%A;50-55 min,20%A;55-60 min,20%→7%A;60-70 min,7%A),the flow rate was 1 m L/min,the detection wavelength was 230 nm for paeoniflorin,240 nm for morroniside and loganin,the column temperature was 30℃,and the injection volume was 10μL.Results The range of paeoniflorin,morroniside and loganin were 29.30-586.00μg/m L(r=0.9998),2.94-58.74μg/m L(r=0.9997),15.29-305.70μg/m L(r=1.0000),respectively.The average recoveries were 97.93%,99.26%,101.45%,and the RSDs were 0.93%,0.60%and 1.40%,respectively(n=6).The contents of paeoniflorin,morroniside and loganin of the 3 batches were in the range of 1.759-1.904 mg/g,0.169-0.188 mg/g and 0.512-0.527 mg/g,respectively.Conclusion This method has high sensitivity and good specificity,which can be used for the content determination of paeoniflorin,morroniside and loganin in Yishen Zhuanggu Pills.
