摘 要:目的基于网络药理学探讨杞菊地黄丸治疗糖尿病眼病的作用机制。方法通过中药系统药理学数据库和分析平台(TCMSP)检索杞菊地黄丸中的药物成分,并筛选出有效活性成分,然后通过TCMSP数据库预测相应靶点,再将预测的靶点输入到UniProt中转换为相应的基因,构建杞菊地黄丸有效成分的靶点网络,再通过PharmGKB数据库检索糖尿病眼病相关基因,构建糖尿病眼病靶点网络,合并杞菊地黄丸有效活性成分的靶点网络与糖尿病眼病的靶点网络,通过筛选取得核心基因团。最后利用DAVID对筛选出的核心基因团进行GO注释和KEGG通路分析。结果杞菊地黄丸有96个有效活性成分,94个靶点,糖尿病眼病有43个相关基因,筛选出核心基因团180个,富集通路主要为甲状腺激素、MAPK、FoxO、PI3K/Akt、胰岛素信号通路。结论杞菊地黄丸的有效活性成分治疗糖尿病眼病可能与甲状腺激素、FoxO与胰岛素的信号通路串话、MAPK、PI3K/Akt等信号通路相关。Objective To explore the mechanism of Qiju Dihuang pill in the treatment of diabetic ophthalmopathy based on network pharmacology.Methods The drug ingredients in Qiju Dihuang pill were searched by traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP),and the effective active ingredients were screened out.Then,the corresponding targets were predicted by TCMSP database,and the predicted targets were input into UniProt and transformed into corresponding genes to construct the target network of effective ingredients of Qiju Dihuang pill.The corresponding genes of diabetic ophthalmopathy was searched by PharmGKB database,and the target network of diabetic ophthalmopathy was constructed.The target network of effective active ingredients of Qiju Dihuang pill and the target network of diabetic ophthalmopathy were combined to obtain the core gene cluster by screening.Finally,DAVID was used for GO annotation and KEGG pathway analysis on the selected core gene cluster.Results The Qiju Dihuang pill had 96 active ingredients and 94 targets.The diabetic ophthalmopathy had 43 related genes.The 180 core gene clusters were screened out.The main enrichment pathways were thyroid hormone,MAPK,FoxO,PI3K/Akt and insulin signaling pathways.Conclusion The active ingredients of Qiju Dihuang pill on diabetic ophthalmopathy may be related to thyroid hormone signaling pathway,crosstalk of signaling pathway of FoxO and insulin,MAPK and PI3K/Akt signaling pathways.
