摘 要:目的研究紫草素在治疗小鼠草酸钙肾结石中的作用。方法用乙醛酸50 mg·kg^-1连续腹腔注射2周建立草酸钙肾结石小鼠模型,腹腔注射生理盐水的小鼠作为对照组。小鼠随机分为3组:对照组、模型组和实验组,每组7只;造模的同时予以给药处理,均灌胃处理,实验组予以紫草素15 mg·kg^-1,对照组和模型组予以同等体积的生理盐水,每天1次,连续给药2周。记录小鼠每天的体重变化,检测小鼠血清肾功能指标;以苏木精-伊红(HE)染色及Von Kossa染色观察小鼠肾组织病理学形态及钙结晶沉积程度。结果紫草素干预下,小鼠体重明显较模型组小鼠高。对照组、模型组和实验组小鼠的肾体比分别为(64.76±3.45),(85.49±5.85),(77.51±4.23)mg·g^-1;血清中肌酐含量分别为(21.22±2.11),(72.86±5.61),(38.80±2.49)μmol·L^-1;尿素氮含量分别为(6.42±0.96),(24.77±1.58),(13.72±1.13)mmol·L^-1;肾组织HE染色评分分别为(0.36±0.09),(3.64±0.62),(1.72±0.15)分;钙结晶沉积评分分别为0,(6.71±0.37),(1.60±0.34)分,实验组和对照组与模型组相比较,差异均有统计学意义(均P<0.05)。结论紫草素可显著减少小鼠草酸钙肾结石的形成,改善小鼠肾功能。Objective To explore the effect of Shikonin on the formation of calcium oxalate stone in mouse kidney.Methods The C57BL/6N mice were given glyoxylate intraperitoneal injection(50 mg·kg^-1)for two weeks to establish calcium oxalate stone mice model.The other mice were treated with normal saline as the control group.The mice were randomly assigned to 3 groups:control group,model group and test group,each group had 7 mice.During the process of establishing the mice model,the test group was given 15 mg·kg^-1 of shikonin,while the control group and the model group were given normal saline.All mice were treated once daily for two weeks.The daily changes in body weight of the mice were recorded,and detected the serum renal function indexes of the mice.The hematoxylin-eosin(HE)staining and Von Kossa staining were used to observe the pathological morphology of kidney tissue and the degree of renal interstitial fibrosis.Results Under the intervention of shikonin,the weight of the mice was significantly higher than that of the model group.The kidney-body ratios of the control group,model group and test group were(64.76±3.45),(85.49±5.85),(77.51±4.23)mg·g^-1;the serum creatinine(Cr)were(21.22±2.11),(72.86±5.61),(38.80±2.49)μmol·L^-1;and the blood urea nitrogen(BUN)were(6.42±0.96),(24.77±1.58),(13.72±1.13)mmol·L^-1.HE staining scores of renal tissue were(0.36±0.09),(3.64±0.62),(1.72±0.15)score;the calcium crystal deposition scores were 0,(6.71±0.37),(1.60±0.34)score.There were significant differences between the control group/test group and the model group(all P<0.05).Conclusion Shikonin can significantly reduce the formation of calcium oxalate stone in mice kidney and improve kidney function in mice.
