摘 要:目的探讨多肽Camel对乳腺癌细胞MCF-7增殖、细胞周期和转录因子(E2F1)基因表达的影响。方法将MCF-7细胞分为2组:实验组与对照组,实验组以Camel处理细胞,对照组加入等量DMEM培养基,分别培养24,48与72 h。用CCK-8法检测乳腺癌细胞MCF-7增殖能力(光密度值),流式细胞仪检测乳腺癌细胞MCF-7的周期阻滞,实时荧光定量-PCR法检测乳腺癌细胞MCF-7中转录因子E2F1基因的相对表达水平。结果于72 h,20μmol·L^-1 Camel组和对照组的细胞增殖率分别为(0.71±0.22)%和(1.12±0.11)%,组间差异有统计学意义(P<0.05)。处理48 h后,10μmol·L^-1 Camel组和对照组的G0/G1期细胞比例分别为(63.70±6.46)%和(53.52±4.12)%;这2组的G2/M期细胞比例分别为(10.05±4.15)%和(17.87±6.23)%;这2组的转录因子E2F1基因相对表达量分别为0.56±0.23和1。上述指标:2组间比较,差异均有统计学意义(均P<0.05)。结论Camel能够抑制MCF-7细胞的增殖,使细胞周期停滞于G0/G1期,且能降低乳腺癌MCF-7细胞中转录因子E2F1基因的表达水平。Objective To investigate the effect of peptide Camel on the proliferation,cell cycle and expression of transcription factor(E2F1)gene in breast cancer cell line MCF-7.Methods MCF-7 cells were divided into two groups:experimental group and control group.The cells in the experimental group were treated with Camel for 24,48 and 72 h,respectively;and the control group were added only with DMEM medium(Dulbecco's modified Eagle medium).The proliferation(OD value)of MCF-7 cells was detected by Cell counting kit-8 assays.The cell cycle arrest of MCF-7 cells was detected by flow cytometry.The relative expression level of E2F1 gene in MCF-7 cells was detected by quantitative real time-PCR.Results The cell proliferation rate at 72 h in 20μmol·L^-1 Camel group and control group were(0.71±0.22)% and(1.12±0.11)%,the difference was statistically significant(P<0.05).After treated 48 h,the cell proportion of G0/G1 phase in 10μmol·L^-1 Camel group and control group were(63.70±6.46)% and(53.52±4.12)%,respectively;and G2/M phase in the two groups were(10.05±4.15)%and(17.87±6.23)%,respectively;the relative expression of E2F1 gene were 0.56±0.23 and 1,respectively;the difference of the factors were statistically significant(all P<0.05).Conclusion Camel could inhibit the proliferation of MCF-7 cells,arrest the cell cycle in the G0/G1 phase,and reduce the expression level of E2F1 gene in breast cancer MCF-7 cells.
