摘 要:目的探讨芹菜素对人卵巢癌SKOV3细胞侵袭、迁移的影响及其相关机制。方法取对数期人卵巢癌SKOV3细胞分为低、中、高剂量实验组及对照组,分别以40,80,160μmol·L^-1芹菜素与等量生理盐水处理,干预48 h。用Transwell小室法检测检测人卵巢癌SKOV3细胞的侵袭能力,用Wound Healing法检测人卵巢癌SKOV3细胞的迁移能力,用四甲基偶氮唑蓝(MTT)比色法检测人卵巢癌SKOV3细胞增殖能力,用蛋白质印迹法检测人卵巢癌SKOV3细胞核因子-κB(NF-κB)和尿激酶型纤溶酶原激活剂(uPA)蛋白表达。结果干预48 h时,对照组与低、中、高剂量实验组人卵巢癌SKOV3细胞的侵袭率分别为(86.43±3.95)%,(62.52±4.11)%,(36.37±3.26)%,(19.21±2.93)%;迁移率分别为(91.22±3.87)%,(72.68±4.21)%,(58.42±3.13)%,(49.02±4.34)%;NF-κB蛋白相对表达量分别为0.91±0.09,0.73±0.11,0.60±0.12,0.49±0.08;uPA蛋白相对表达量分别为0.96±0.04,0.68±0.13,0.52±0.09,0.41±0.07。低、中、高剂量实验组分别与对照组比较,差异均有统计学意义(均P<0.05)。结论芹菜素可有效抑制人卵巢癌SKOV3细胞的增殖、侵袭和迁移能力,其作用机制可能与下调人卵巢癌SKOV3细胞内NF-κB和uPA蛋白表达相关。Objective To investigate the effect of apigenin on the invasion and migration of human ovarian cancer SKOV3 cells and its related mechanisms.Methods Human ovarian cancer SKOV3 cells in logarithmic phase were divided into the exp-L/-M/-H group and the control group,and treated with 40,80,and 160μmol·L^-1 apigenin and the same amount of normal saline,and intervened for 48 h.Transwell chamber method was used to detect the invasion ability of human ovarian cancer SKOV3 cells;Wound Healing method was used to detect the migration ability of human ovarian cancer SKOV3 cells;The methyl thiazolyl tetrazolium(MTT)method was used to detect the proliferation of human ovarian cancer SKOV3 cells;Western blot was used to detect nuclear factor-κB(NF-κB)and urokinase-type plasminogen activator(uPA)protein expression in human ovarian cancer SKOV3 cells.Results At 48 h of intervention,the invasion rates of human ovarian cancer SKOV3 cells in the control and exp-L/-M/-H groups were(86.43±3.95)%,(62.52±4.11)%,(36.37±3.26)%,and(19.21±2.93)%;The migration rates were(91.22±3.87)%,(72.68±4.21)%,(58.42±3.13)%,(49.02±4.34)%;The relative expression levels of NF-κB protein were 0.91±0.09,0.73±0.11,0.60±0.12,0.49±0.08;The relative expression levels of uPA protein were 0.96±0.04,0.68±0.13,0.52±0.09,and 0.41±0.07.There were statistically significant differences in the above indicators between the control group and the exp-L/-M/-H group(all P<0.05).Conclusion Apigenin can effectively inhibit the proliferation,invasion and migration of human ovarian cancer SKOV3 cells,and its mechanism may be related to the down-regulation of NF-κB and uPA protein expression in human ovarian cancer SKOV3 cells.
