摘 要:目的探讨吴茱萸碱对宫颈癌HeLa细胞增殖与凋亡的影响及可能的机制。方法体外培养宫颈癌细胞株HeLa,低、中、高剂量实验组及对照组分别以浓度为25,50,100μg·mL-1吴茱萸碱和等量生理盐水干预,分别干预12、24、48 h。用光学倒置显微镜观察各组HeLa细胞形态变化;用四甲基偶氮唑蓝(MTT)法测定HeLa细胞的增殖抑制率;用Hoechst 33258染色检测HeLa细胞的凋亡情况;用蛋白质印迹法检测HeLa细胞谷胱甘肽S转移酶P1(GSTP1)、磷酸化-c-Jun氨基末端激酶(p-JNK)蛋白的表达水平。结果干预48 h时,对照组与低、中、高剂量实验组HeLa细胞的增殖抑制率分别为0%,(43.45±4.18)%,(56.86±4.37)%和(74.69±5.11)%;凋亡率分别为(13.42±2.89)%,(27.84±3.11)%,(54.16±4.12)%,(80.21±5.34)%;HeLa细胞GSTP1蛋白相对表达量分别为0.95±0.05,0.72±0.11,0.53±0.12,0.16±0.08;p-JNK蛋白相对表达量分别为0.11±0.06,0.33±0.14,0.47±0.09,0.84±0.12。以上指标,低、中、高剂量实验组分别与对照组比较,低、中、高剂量实验组比较,差异均有统计学意义(均P<0.05)。结论吴茱萸碱可抑制宫颈癌HeLa细胞增殖,促进其凋亡,抑制GSTP1、激活JNK通路可能是其发挥抑制HeLa细胞增殖,诱导凋亡的作用机制。Objective To investigate the effect of evodiamine on the proliferation and apoptosis of cervical cancer HeLa cells and its possible mechanism.Methods The cervical cancer cell line HeLa were cultured in vitro,and the exp-L/-M/-H group and the control group were treated with 25,50,100μg·mL-1 of evodiamine and the same amount of normal saline,and intervened for 12,24,48 h.The optical inverted microscope was used to observe morphological changes of HeLa cells;Proliferation inhibition rate of HeLa cells was measured by methyl thiazolyl tetrazolium(MTT)method;Hoechst 33258 staining was used to detect the apoptosis of HeLa cells;the expression levels of glutathione S-transferase P1(GSTP1)and phosphorylated-c-Jun amino-terminal kinase(p-JNK)protein in HeLa cells were detected by Western blotting.Results After 48 h of intervention,the proliferation inhibition rates of HeLa cells in the control group and exp-L/-M/-H group were 0%,(43.45±4.18)%,(56.86±4.37)%,and(74.69±5.11)%,respectively;the apoptosis rates were(13.42±2.89)%,(27.84±3.11)%,(54.16±4.12)%,and(80.21±5.34)%;The relative expression levels of GSTP1 protein in HeLa cells were 0.95±0.05,0.72±0.11,0.53±0.12,0.16±0.08;The relative expression levels of p-JNK protein were 0.11±0.06,0.33±0.14,0.47±0.09,and 0.84±0.12,all with significant differences(all P<0.05).Conclusion Evodiamine can inhibit the proliferation of cervical cancer HeLa cells,promote its apoptosis,inhibit GSTP1,and activating the JNK pathway may be its mechanism of inhibiting HeLa cell proliferation and inducing apoptosis.
