摘 要:目的筛选实验性关节炎发病过程中与辅助性T细胞17(Th17)分化信号通路有关的差异表达信使核糖核酸(mRNA)并探讨实验性关节炎可能的发病机制。方法按照随机数字表法将大鼠随机分为2组:对照组和实验组,每组3只。实验组大鼠左后足皮下注射弗氏完全佐剂0.1 mL制备关节炎模型,对照组予以等量0.9%NaCl。造模后第21天,深度麻醉下取2组大鼠滑膜组织,用Array star大鼠全基因组芯片来检测其mRNAs表达情况,并以Fold change值>2.0,P<0.05筛选差异表达mRNA;进行信号通路分析,筛选与Th17细胞分化有关的mRNA。结果与对照组相比,实验组共筛选出4547个差异表达mRNA,并获得138条与差异基因相关的信号通路。与Th17细胞分化相关通路是富集分数第3位的信号通路,其中有36个基因均为上调表达;Th17细胞的标志物IL-17mRNA差异表达倍数为3.07,Treg细胞的标志物Foxp3 mRNA差异表达倍数为23.02。结论Th17细胞分化相关基因在实验性关节炎发病过程中存在差异表达。Objective To screen the differential expression messenger ribonucleic acid(mRNA)related to T helper cell 17(Th17)differentiation signal pathway in the pathogenesis of experimental arthritis and to explore the possible mechanism of experimental arthritis.Methods Rats were randomly divided into two groups according to random number table:control group and experimental group,with 3 rats in each group.The rats in the experimental group were subcutaneously injected with 0.1 mL Freund’s complete adjuvant to prepare an arthritis model.The control group was given an equal amount of saline.On day 21,the synovial tissues of rats in two groups were taken under deep anesthesia,and the Array star rat whole genome chip was used to detect differentially expressed messenger ribonucleic acid(mRNA).The differential expression mRNA was screened with Fold change value>2.0 and P-value<0.05.The signal pathway analysis was performed to screen the mRNA associated with Th17 cell differentiation.Results Compared with the control group,a total of 4547 differentially expressed mRNAs were selected in the experimental group,and 138 signal pathways related to differential genes were obtained.Th17 cell differentiation-related pathway is the third signal pathway based on enrichment score,and 36 genes were all up-regulated genes.Among them,the differential expression fold of Th17 cell marker IL-17 mRNA is 3.07,the differential expression fold of Treg cell marker Foxp3 mRNA is 23.02.Conclusion Th17 cell differentiation-related genes are differentially expressed in experimental arthritis.
