摘 要:目的探讨番红花提取物对骨质疏松大鼠股骨骨折后愈合速度和炎症反应的影响。方法选取SPF级6~8月龄的雄性大鼠40只,按照随机数字表法分为空白组、模型组、低剂量实验组和高剂量实验组,每组10只;空白组不做处理,模型组、低剂量实验组和高剂量实验组予以灌胃维甲酸70 mg·kg^-1,每日1次,共14 d,导致大鼠骨质疏松;4组大鼠均采用闭合骨折模型法造成大鼠右侧股骨中段骨折;造模成功后,空白组和模型组常规标准饲养;低剂量实验组予以灌胃番红花提取物5 mg,每日1次;高剂量实验组予以灌胃番红花提取物10 mg,每日1次;低、高剂量实验组均连续给药14 d;用酶联免疫吸附试验测定大鼠尾静脉血中肿瘤坏死因子-α(TNF-α),用试剂盒测定谷胱甘肽(GSH)水平;检测骨钙素(OC);于骨折愈合后处死大鼠,取造模的股骨,用微型CT测定骨小梁密度,记录大鼠骨折愈合时间。结果治疗后,空白组、模型组、低剂量实验组、高剂量实验组血清TNF-α水平分别为(17.71±3.23),(33.21±5.45),(27.37±4.26),(21.70±3.73)pg·mL^-1,上述4组血清GSH水平分别为(68.43±6.51),(48.72±8.69),(57.13±9.95),(62.51±8.77)μmol·L^-1,上述4组血清OC水平分别为(8.04±0.81),(5.59±0.57),(5.82±0.66),(6.33±0.53)ng·mL^-1,上述4组骨小梁密度分别为(793.40±34.75),(533.58±34.43),(633.87±42.74),(699.29±34.90)mg·(mm^3)^-1,上述4组骨折愈合速度分别为上述4组骨折愈合速度分别为(9.13±1.77),(14.99±2.75),(12.77±2.56),(10.42±1.58)周;上述指标模型组与空白组比较,差异均有统计学意义(均P<0.05);低、高剂量实验组与模型组比较,差异均有统计学意义(均P<0.05);低剂量实验组与高剂量实验组比较,差异均有统计学意义(P<0.05);结论番红花提取物能够促进骨质疏松大鼠的骨折愈合,减轻大鼠体内炎症反应和氧化应激状态。Objective To investigate the effect of saffron extract on the healing speed and inflammation of osteoporotic rats after femoral fracture.Methods Forty male SPF grade 6-8 months old rats were selected and divided into blank group,model group,test-L group and test-H group according to random number table method,10 rats in each group;The blank group was not treated,and the model group,the test-L group and the test-H group were intragastrically fed with retinoic acid 70 mg·kg^-1 for 14 days,once daily,resulting in osteoporosis in rats;All four groups of rats used the closed fracture model to cause the fracture of the right middle femur of the rat;After successful modeling,the blank group and the model group were given standard diet;the test-L group was given oral administration of saffron extract 5 mg once daily;the test-H group was given oral administration of saffron extract 10 mg once daily.All groups were treated for 14 days;Determination of tumor necrosis factor-α(TNF-α)in rat tail vein blood was conducted using enzyme-linked immunosorbent assay;glutathione(GSH)level was measured by kit;osteocalcin(OC)was measured;After the fracture was healed,the rats were killed,the modeled femur was taken,and the density of the trabecular bone was measured by micro CT;The fracture healing rates were recorded.Results After treatment,the serum TNF-αlevels in the blank group,model group,test-L group,and test-H group were(17.71±3.23),(33.21±5.45),(27.37±4.26),(21.70±3.73)pg·mL-1;The serum GSH levels of the above four groups were(68.43±6.51),(48.72±8.69),(57.13±9.95),(62.51±8.77)μmol·L^-1;The serum OC levels of the above four groups were(8.04±0.81),(5.59±0.57),(5.82±0.66),(6.33±0.53)ng·mL^-1;The trabecular bone density of the above four groups were(793.40±34.75),(533.58±34.43),(633.87±42.74),(699.29±34.90)mg·(mm^3)^-1;The fracture healing rates of the above four groups were(9.13±1.77),(14.99±2.75),(12.77±2.56),(10.42±1.58)weeks.Compared the above index model group and the blank group,the differences we
