摘  要:目的:探讨高强度间歇性训练(HIIT)对中年大鼠心肌细胞凋亡与增殖相关蛋白表达的影响。方法:选取9月龄wistar大鼠70只。随机分为基础值组(B组),4周安静对照组(Q1组)、4周中等强度运动组(M1组)、4周HIIT组(H1组)和8周安静对照组(Q2组)、8周中等强度运动组(M2组)、8周HIIT组(H2组),每组10只。安静对照不进行运动。中等强度运动组:大鼠进行跑台训练。实验运动前进行VO2max测试,确定训练方案平均运动强度约为60%VO2max,每天训练1次,每次50 min,每周5 d。HIIT运动组:实验运动前进行VO2max测试,并根据测试结果及大鼠达到最大摄氧量时的跑台运动速度,确定HIIT运动方案进行为期8周的训练,每天训练1次,每次50 min,每周训练5 d。训练结束后,测定最大摄氧量和体重;基础值组大鼠在首次最大摄氧量测试结束后24 h取材,.安静对照组和运动干预组大鼠在干预第4、8周最大摄氧量测试结束后24 h进行取材,心脏称重后以心脏重量与体重比值计算心肌指数(心脏重量/体重,mg/g),采用Western Blotting检测心肌细胞凋亡与增殖相关蛋白:Bcl-2相关X蛋白(Bax)、B细胞淋巴瘤-2基因(Bcl-2)、活化半胱天冬蛋白酶-3(Cleaved Caspase-3)、活化聚腺苷二磷酸核糖聚合酶-1(Cleaved PARP-1)、增殖细胞核抗原(PCNA)。结果:干预8周后从安静组到中等强度运动组再到HIIT组,心肌指数呈现上升趋势,其中HIIT组显著高于安静组(P<0.05);干预8周后HIIT组VO2max相比干预前显著提高(P<0.01),与同周安静组相比显著提高(P<0.05);Bax蛋白表达:H2组与Q2组、M2组相比显著提高23.5%、22.2%(P<0.01,P<0.05)。Bax/Bcl-2比值:干预8周后各组总体呈上升趋势,H2组与H1组、Q2组、M2组相比显著提高53.8%、56.8%、34.7%(P<0.01,P<0.01,P<0.05)。Cleaved Caspase-3蛋白表达:干预8周后各组总体呈上升趋势,H2组与H1组、Q2组、M2组相比显著提高24.2%、33.7%、18.3%(P<0.05)。Cleaved PARP-1蛋白表达:H2组与Objective To explore the effect of high intensity interval training(HIIT)on apoptosis and proliferation-related protein expression of cardiomyocytes of middle-aged rats.Methods Seventy 9-month-old Wistar rats were selected and randomly divided into a basic value group(Group B),4-week control group(Group Q1),4-week moderate intensity training group(Group M1),4-week HIIT group(Group H1),8-week control group(Group Q2),8-week moderate intensity training group(Group M2)and 8-week HIIT group(Group H2),each of 10.Rats in the control group didn’t do any exercise.In the moderate-and high-intensity training groups,rats underwent 50-minute running on treadmills with the average exercise intensity of 60% and 100% VO2max respectively 5 days a week.After the training,the maximum oxygen uptake and body weight were measured.The rats of Group B were killed and collected their hearts 24 hours after the first maximum oxygen uptake test,while the other groups were done after 4 or 8 week of intervention.After the heart was weighed,the myocardial index(heart weight/body weight,mg/g)was calculated based on the ratio of the heart weight to body weight,and Western blotting was used to detect the myocardial apoptosis and proliferation of related proteins including Bcl-2 related X Protein(Bax),B cell lymphoma-2 gene(Bcl-2),activated caspase-3(cleaved caspase-3),activated poly(ADP-ribose)polymerase-1(cleaved PARP-1)and proliferating cell nuclear antigen(PCNA).Results After 8 weeks of intervention,the myocardial index of all groups showed an increasing trend,with that of HIIT groups significantly higher than the control group(P<0.05).After the intervention,the average VO2max of Group H2 was significantly higher than before the intervention(P<0.01)and Group Q2 at the same time point(P<0.05).After the intervention,the average Bax expression of Group H2 increased significantly by 23.5% and 22.2% respectively compared with Group Q2 and Group M2(P<0.01,P<0.05),and the average Bax/Bcl-2 ratio was found to be higher in all groups,with that of Gro
