摘 要:目的比较EV71与柯萨奇病毒A16病毒载量与人横纹肌瘤细胞(RD)病变效应的关系。方法测定多株EV71和柯萨奇病毒A16毒株的半数培养感染量(TCID50);使用real-time PCR建立标准曲线,测定两类毒株分离液的病毒载量与每个TCID50单位相应的病毒载量。结果EV71和柯萨奇病毒A16毒株分离液病毒载量差异无统计学意义(t=-1.27,P=0.21);EV71毒株每个TCID50单位相应的核酸载量显著低于柯萨奇病毒A16毒株(t=4.29,P<0.01)。结论EV71与柯萨奇病毒A16感染人横纹肌瘤细胞产生的病毒颗粒总量无差异,正常病毒与缺陷病毒颗粒的相对量有明显差异,这可能是2种病毒致病性差异的原因之一。Objective To compare the relationship between the cytopathic effect and virus load of EV71 and CoxA16 cultured in rhabdomyoma(RD)cells.Methods TCID50 of several EV71 and CoxA16 strains were detected.Real-time PCR was used to establish the standard curve to determine the viral load of the two strains isolates and the corresponding viral load of each TCID50 unit.Results There was no statistical significance on the difference in viral load between EV71 and Coxsackie virus A16 isolates(t=-1.27,P=0.21).The nucleic acid load per TCID50 unit of EV71 was significantly lower than that of Coxsackie Virus A16(t=4.29,P<0.01).Conclusion There is no difference on the total amount of virus particles produced by EV71 and Coxsackie virus A16 infected human rhabdomyoma cells,and the relative amount of normal virus and defective virus particles were significantly different,which may be one of the reasons for the difference in pathogenicity of the two viruses.
