摘　　要：目的探究HLA-G 3'UTR 14 bp基因多态性与先兆流产的相关性。方法选取确诊为先兆流产的患者264例作为疾病组,正常妊娠妇女93例作为对照组,进行疾病对照研究。离心柱法提取全血DNA基因,PCR扩增HLA-G 3'UTR区域的DNA,Sanger测序法对基因进行测序分型,SHEsis软件统计分析基因型和等位基因频率,进行假定遗传模式分析。结果HLA-G 3'UTR 14 bp Ins/Del位点等位基因Ins在病例组的分布高于对照组,与Del比较差异有统计学意义(P<0.05)。3种基因型在2组间的分布差异无统计学意义(P>0.05)。不同遗传模式下,14 bp基因型在2组间的分布差异无统计学意义(P>0.05)。结论14 bp Ins等位基因是先兆流产的风险因素,HLA-G 3'UTR 14 bp基因多态性与先兆流产有一定相关性。Objective To explore the correlation of HLA-G 3'UTR 14 bp gene polymorphism and threatened abortion.Methods Case-control studies were conducted,including the patients diagnosed as threatened abortion as the case group and normal pregnant women as the control group.Whole blood DNA genes were extracted by centrifuge column method,and DNA was amplified by PCR.Genes were sequenced and classified by Sanger sequencing method.Genotypes and allele frequency were statistically analyzed by SHEsis software,and assumed genetic model was analyzed,subsequently.Results The distribution of ALLelic Ins of HLA-G 3'UTR 14 bp Ins/Del locus in the case group was higher than that in the control group,and the difference was statistically significant(P<0.05).There was no statistical significance on the difference in the distribution of three genotypes between the two groups(P>0.05).Under different genetic patterns,there was no statistical significance on the difference in the distribution of 14 bp genotype between the two groups(P>0.05).Conclusion 14 bp Ins allele is a risk factor of threatened abortion,so HLA-G 3'UTR 14 bp gene polymorphism and threatened abortion may have relevance.
