摘 要:目的研究筋骨草对慢性气道炎症小鼠的治疗作用及其对肠道菌群结构的调节作用。方法选取SPF级ICR小鼠60只,随机分为正常对照组(C)、慢性气道炎症模型组(M)、筋骨草低剂量治疗组(L)、筋骨草中剂量治疗组(MI)、筋骨草高剂量治疗组(H)、阳性治疗组(P)。烟熏法建立慢性气道炎症模型后给药28 d,检测肺泡灌洗液中TNF-α、IL-10、IL-6水平及其他指标,收集各组小鼠粪便,进行16SrRNA测序,分析治疗后肠道菌群的变化。结果慢性气道炎症模型组TNF-α、IL-6明显升高,IL-10降低,筋骨草治疗后IL-10升高,TNF-α、IL-6水平降低。16SrRNA测序结果显示筋骨草治疗后小鼠肠道菌群的多样性和丰富度均显著增加,各组小鼠肠道菌群的差异分析结果显示筋骨草治疗后肠道中拟杆菌比例显著升高。结论筋骨草治疗小鼠慢性气道炎症疗效显著,其可能通过调节肠道菌群结构,增加拟杆菌比例,改善炎症反应,发挥治疗作用。Objective To study Jingucao(Ajuga)’s therapeutic effect on chronic airway inflammation and its regulation on intestinal flora in mice.Methods Sixty SPF ICR mice were randomly divided into normal control group(C),chronic airway inflammation model group(M),low-dose treatment group(L),middle-dose treatment group(MI),high-dose treatment group(H),and the positive treatment group(P).Smoke method was used to establish a chronic airway inflammation model and then sick mice were treated for 28 days.The levels of TNF-α,IL-10 and IL-6 in bronchoalveolar lavage fluid and other indicators were detected.The feces of each group were collected and 16 SrRNA sequencing was performed to analyze the changes of intestinal flora after treatment.Results In the chronic airway inflammation model group,the levels of TNF-αand IL-6 were significantly increased.However,the level of IL-10 was decreased.After treatment with Jingucao(Ajuga),the level of IL-10 was increased,and the levels of TNF-αand IL-8 were decreased.The results of 16 SrRNA showed that the diversity and abundance of intestinal flora in mice treated with Jingucao(Ajuga)increased significantly.The results of differential analysis of intestinal flora in each group showed that the proportion of Bacteroides in the intestinal tract was significantly increased after treatment with Jingucao(Ajuga).Conclusion Jingucao(Ajuga)can improve chronic airway inflammation in mice,and can regulate the structure of intestinal flora and increase the proportion of Bacteroides.
