摘 要:目的讨论麻黄定喘汤对支气管哮喘的作用机制,其机制是否与WNT-5A有关。方法将50只BABL/c小鼠随机分成正常组、模型组、麻黄定喘汤低剂量组、麻黄定喘汤高剂量组、地塞米松组(10只/组)。通过致敏(OVA、氢氧化铝),激发(OVA、NS)建立小鼠哮喘模型。末次激发24 h后解剖小鼠取肺泡灌洗液(BALF)和肺组织。左肺组织进行苏木精-伊红(HE)、PAS染色观察肺组织中病理学改变。采用免疫吸附(ELISA)法检测BALF中炎性细胞种类及炎性细胞因子白细胞介素-4(IL-4)、白细胞介素-5(IL-5)、白细胞介素-13(IL13)、干扰素-γ(IFN-γ)的含量。结果与正常组相比,模型组小鼠的BALF中炎性细胞数增多,IL-4、IL-5、IL-13水平明显增高(P<0.01)。与模型组相比,麻黄定喘汤高、低剂量组和地塞米松组小鼠的BALF中的炎性细胞数、IL-4、IL-5、IL-13水平。结论麻黄定喘汤可能通过WNT-5A抑制支气管哮喘气道炎症的发生。Objective To discuss the mechanism of Chao medicine Mahuang Dingchuan Decoction on bronchial asthma and whether its mechanism is related to WNT-5 A.Methods Fifty BABL/c mice were randomly divided into normal group,model group,low dose group of Mahuang Dingchuan Decoction,high dose group of Mahuang Dingchuan Decoction and dexamethasone group(10 mice/group).The asthma model was established by sensitization(OVA,aluminum hydroxide)and challenge(OVA,NS).After 24 hours of the last challenge,the mice were dissected to take alveolar lavage fluid(BALF)and lung tissue.Pathological changes in left lung tissue were observed by hematoxylin-eosin(HE)and PAS staining.We detected inflammatory cell types and inflammatory cytokines interleukin-4(IL-4),interleukin-5(IL-5)and interleukin-13(IL-13)in BALF and the content of interferon-γ(IFN-γ)by immunosorbent assay(ELISA).Conclusion Compared with those of the normal group,the number of inflammatory cells and the levels of IL-4,IL-5 and IL-13 in the BALF of the model group increased significantly(P<0.01).Compared with those of the model group,the number of inflammatory cells,IL-4,IL-5,IL-13 and WNT-5 A in BALF were significantly higher in the high dose and low dose groups of Mahuang Dingchuan Decoction and dexamethasone group.reduce.Conclusion Mahuang Dingchuan Decoction may inhibit the occurrence of airway inflammation in bronchial asthma by WNT-5 A.
