摘  要:目的探究白细胞介素1(IL-1)是否通过Wnt/β-catenin信号通路调控椎间盘退变。方法将30只C57bl/6小鼠随机分成5组,分别为正常组,IL-1低、中、高剂量干预组(0.04、0.2、1μg/10g),椎间盘退变模型组。IL-1干预组采用腹腔注射正常小鼠IL-1,1次/d;采用诱导站立法制备椎间盘退变小鼠模型。X射线荧屏下观察各组小鼠腰椎结构变化,Micro-CT分析各组小鼠腰椎显微结构的改变,HE染色观察各组小鼠椎间盘病理学改变,免疫组化和Westerm blot检测各组小鼠椎间盘内相关蛋白的表达情况,采用qRT-PCR技术检测相关基因的表达情况。结果与正常组比较,IL-1千预组和椎间盘退变模型组腰椎结构明显发生退变,骨密度(BMC/TV)及骨小梁数量(Tb.N)显著降低,腰椎盘病理学改变程度明显增加(P<0.05)。并且与正常组比较,IL-1干预组和椎间盘退变模型组小鼠椎间盘内IL-1和β-catenin的蛋白及基因表达水平显著升高,Ⅱ型胶原(CollagenⅡ)和蛋白聚糖(Versican)的蛋白及基因表达水平显著降低(P<0.05)。结论IL-1能够通过Wnt/β-catenin信号通路调控椎间盘退变。Objective To investigate whether interleukin 1(IL-1)regulates intervertebral disk degeneration through the Wnt/β-catenin signaling pathway.Methods Thirty C57bl/6 mice were randomly divided into five groups:normal group,low,medium and high dose IL-1 intervention group(0.04,0.2,1μg/10g),and disk degeneration model group.The IL-1 group received intraperitoneal injection of normal mice IL-1 once a day.The model of intervertebral disk degeneration was prepared by induced standing.X-ray was used to observe the changes in the lumbar spine structure of mice in each group,Micro-CT was used to analyze the changes in the lumbar spine microstructure of mice in each group,HE staining was used to observe the pathological changes in the intervertebral discs of mice in each group,immunohistochemistry and Western blot were used to detect the expression of related proteins in each group,and qRT-PCR was used to detect the expression of related genes.Results Compared with the normal group,the IL-1 intervention group and the model group showed significant degeneration in lumbar structure,signifcantly decreased BMC/TV and Tb.N,and significantly increased pathological changes in lumbar discs.Compared with normal group,the protein and mRNA levels ofIL-1 andβ-cateninn in IL-1 intervention group and model group were significantly increased.CollagenⅡand Versican were significantly decreased(P<0.05)。Conclusion IL-1 regulates intervertebral disk degeneration through the Wnt/β-catenin signaling pathway.
