摘  要:以蔗糖和谷氨酸为原料,聚乙烯亚胺为钝化剂,合成一种表面带正电荷的、N掺杂的阳离子碳点;通过高分辨透射电子显微镜、傅里叶变换红外光谱、X射线光电子能谱、紫外光谱和荧光光谱等技术手段对制备的阳离子碳点的形貌、表面官能团以及光学特性进行表征;利用荧光光谱考察阳离子碳点对人体纤维原蛋白的猝灭机制;联合同步荧光和圆二色谱,研究阳离子碳点对人体纤维原蛋白空间构象的影响。结果表明:阳离子碳点粒径约为2.2 nm,呈规则圆球形,Zeta电位为+28.7 mV,且表面存在羰基、羧基、羟基和胺基等官能团;在模拟生理条件下,当阳离子碳点浓度为0~2.0μmol/L时,人体纤维原蛋白的荧光随阳离子碳点浓度的增大呈线性静态猝灭,两者以静电作用力结合;阳离子碳点与人体纤维原蛋白的色氨酸残基作用更强,阳离子碳点浓度大于1.0μmol/L时会引起人体纤维原蛋白解螺旋。A kind of nitrogen doped cationic carbon dots with positive surface charges were synthesized taking sucrose and glutamic acid as materials and polyethylenimine as the passivator.The morphology,surface functional groups,and optical properties of prepared cationic carbon dots were characterized by using high resolution transmission electron microscopy,Fourier transform infrared spectroscopy,X-ray photoelectron spectroscopy,ultraviolet visible spectroscopy,fluorescence spectroscopy,and so on.The quenching mechanism of cationic carbon dots on human fibrinogen was measured by using the fluorescence spectroscopy.The effect of cationic carbon dots on spatial conformation of human fibrinogen were demonstrated using both synchronous fluorescence and circular dichroism spectra.The results show that cationic carbon dots are regularly spherical with particle size of about 2.2 nm and Zeta potential of+28.7 mV.The surface is linked by functional groups of carbonyl, carboxyl, hydroxyl, amine, and so on. Under simulated physiological conditions, when the concentration of cationic carbon dots ranges from 0 to 2.0 μmol/L, the fluorescence of human fibrinogen quenches staticly and linearly with the increase of cationic carbon dots concentration. The cationic carbon dots interact with human fibrinogen through electrostatic force. The tryptophan residues of human fibrinogen are more easily affected by cationic carbon dots. The concentration of cationic carbon dots over 1.0 μmol/L induces the α -helix structure of human fibrinogen to unfold.
