摘　　要：目的动态观察皮肤再生医疗技术(moist exposed burn therapy/moist exposed burn ointment,MEBT/MEBO)对大鼠慢性难愈合创面组织中BMP-4表达的影响。方法通过随机数字表将90只SPF雄性大鼠随机分为MEBO组、阳性药组(贝复济组)、模型组、急创组和空白组,构建大鼠慢性难愈合创面模型后,MEBO组创面局部涂用湿润烧伤膏,贝复济组创面局部涂用贝复济,模型组和急创组局部涂用生理盐水,于治疗后第3 d、7 d、14 d应用Western blotting检测大鼠创面组织中BMP-4表达情况,于治疗后第14 d,应用Masson染色,观察创面局部病理学形态。结果(1)除空白组外,其余各组BMP-4的表达水平均呈先升高后降低的趋势(P<0.01),同一时间,MEBO组及阳性药组创面组织中BMP-4的表达水平均高于模型组(P<0.05);(2)MEBO组及阳性药组创面组织病理学形态优于模型组。结论MEBT/MEBO能有效影响大鼠创面组织中BMP-4表达水平,从而影响慢性难愈合创面修复的质量。Objective To dynamically observe the effect of moist exposed burn therapy/moist exposed burn ointment(MEBT/MEBO)on the expression of BMP-4 in chronic refractory wound tissues of rats.Methods A total of 90 SPF male rats were randomly divided into the MEBO group,the positive drug group(rb-bFGF)group,the model group,the acute wound group and the blank group by random number table.After the establishment of chronic refractory wound models in rats,wounds in MEBO group were locally coated with MEBO,rb-bFGF Group with rb-bFGF,and the model group and acute wound group with normal saline.On the 3 rd,7 th and 14 th day after treatment,Western blotting was used to detect the expression of BMP-4 in wound tissues of rats.On the 14 th day after treatment,Masson staining was applied and the local pathological morphology of the wound was observed.Results(1)Except for that of the blank group,the expressions of BMP-4 in other groups showed a trend of increasing at first and then decreasing(P<0.01).At the same time,the expressions of BMP-4 in wound tissue of the MEBO group and the positive drug group were higher than that of the model group(P<0.05).(2)The histopathological morphology of wounds of the MEBO group and the positive drug group was superior to that of the model group.Conclusion MEBT/MEBO can effectively affect the expression of BMP-4 in wound tissues of rats,thus improving the repair quality of chronic refractory wound.
