摘  要:[目的]为了研究除草剂拿捕净对谷子愈伤组织细胞抗坏血酸-谷胱甘肽(AsA-GSH)循环的影响,探究不同感抗性谷子品种对拿捕净胁迫的生理响应,为谷子抗除草剂胁迫机制提供理论参考。[方法]以拿捕净敏感品种晋谷21和抗性品种豫谷35、冀谷42为试验材料,设置拿捕净浓度分别为0、0.5、1、2、3 mL·L-15个水平,观察拿捕净对3种谷子种子萌发、愈伤诱导和生长的影响,测定AsA-GSH循环中抗坏血酸过氧化物酶(APX)、谷胱甘肽还原酶(GR)活性,还原型抗坏血酸(AsA)、脱氢型抗坏血酸(DHA)、总抗坏血酸(AsA+DHA)、还原型谷胱甘肽(GSH)含量变化,分析其对拿扑净胁迫不同程度的响应。[结果]随着拿捕净浓度的升高,谷子的萌发、愈伤组织的诱导以及愈伤组织的生长受到抑制,与敏感品种晋谷21相比,抗性品种豫谷35和冀谷42APX、GR的活性及AsA、DHA、GSH含量均较高。[结论]提高拿捕净胁迫浓度明显抑制3个品种谷子的萌发、愈伤组织的诱导和生长,2个抗性品种豫谷35和冀谷42各抗性指标变化趋势不完全一致,但总体高于敏感品种晋谷21,表明其可通过提高保护酶活性和抗氧化剂含量来响应拿捕净胁迫。[Objective]The objective of this experiment was to investigate the effect of herbicide sethoxydim on the ascorbic acidglutathione(AsA-GSH)circulation in callus of foxtail millet,explore the physiological response of different resistant foxtail millet varieties to sethoxydim stress.The result could provide theoretical reference for the stress mechanism of foxtail millet herbicide resistance.[Methods]This experiment was conducted with Jingu 21(sensitive to sethoxydim),Yugu 35 and Jigu 42(resistant to sethoxydim)as materials with five dosages(0、0.5、1、2、3 mL·L-1)of sethoxydim,the effect of sethoxydim on germination,callus induction and growth of three species of foxtail millet were observed,there are indicators were determined in AsA-GSH cycle,including the activities of aseorbateperoxidase(APX),glutathione reductase(GR),and the contents of ascorbic acid(AsA),docosahexaenoic acid(DHA),total ascorbic acid(AsA+DHA),andglutathione(GSH),and their response to sethoxydim stress was analyzed to varying degrees.[Results]The results showed that with the increase of sethoxydim concentration,the germination,callus induction and callus growth of foxtail millet were inhibited,and compared with sensitive variety Jingu 21,the activity of APX、GR and the content of AsA、DHA、GSH in resistant varieties Yugu 35 and Jigu 42 were higher.[Conclusion]Increasing the concentration of sethoxydim stress obviously inhibited the germination and growth of three varieties of foxtail millet.The change trend of resistance indexes of two resistant varieties Yugu 35 and Jigu 42 was not exactly consistent,but they were higher than the sensitive variety Jingu 21,which indicates that the resistant varieties could resist sethoxydim stress by increasing the activity of protective enzyme and antioxidant content.
