摘  要:目的通过分离、鉴定并培养大鼠卵巢颗粒细胞,筛选出能够抑制雷公藤多苷对大鼠卵巢颗粒细胞损伤的药物。方法收集21~23 d龄的SD大鼠卵巢颗粒细胞,经FSRH免疫荧光鉴定。设置空白组、模型组、5种药材(过路黄、藤茶、淫羊藿、黄芪、当归)与雷公藤多苷的样品组,采用CCK8法检测比较各组卵巢颗粒细胞的存活率;ELISA法测定各组卵巢颗粒细胞所释放雌二醇及孕酮的量。结果雷公藤多苷干预大鼠卵巢颗粒细胞后,细胞存活率明显降低,雌二醇和孕酮的释放量明显减少。分别加入过路黄、藤茶、淫羊藿、当归、黄芪配伍,结果显示雷公藤多苷与黄芪配伍后其卵巢颗粒细胞细胞毒性显著减小,雌二醇和孕酮的释放明显增加。结论黄芪能够逆转雷公藤多苷对SD大鼠卵巢颗粒细胞的细胞毒性,增加雌二醇和孕酮的释放。Objective To isolate,identify and culture rat ovarian granulosa cells,and to screen drugs to inhibit the damage of triptolide to rat ovarian granulosa cells.Methods Ovary granulosa cells were collected from 21-23 day-old SD rats and identified by FSRH cellular immunofluorescence.Ovary granulosa cells were randomly divided into 7 groups which received complete medium,tripterygium glycosides,tripterygium glycosides together with scutellaria,ampelopsis grossedentata,epimedium,astragalus and angelica extact.The proliferation of ovary granulosa cells was detected by CCK8,and the estradiol and progesterone were determined by ELISA.Results Tripterygium glycosides significantly reduced the survival rate of ovarian granulosa cells,and estradiol and also reduced progesterone.However,when used together with astragalus,the ovarian granulosa cell cytotoxicity of tripterygium glycosides was significantly reduced,while that of estradiol and progesterone was significantly increased.Conclusion Astragalus can reverse the cytotoxicity of tripterygium glycosides in ovarian granulosa cells of SD rats and increase the release of estradiol and progesterone.
