摘 要:目的评价新疆沙枣叶水提取物(EALs)体内外抗肿瘤活性。方法采用MTT法测定EALs的体外抑瘤作用,计算半数抑制浓度(IC50)。通过建立S180荷瘤小鼠模型,观察不同浓度EALs对荷瘤小鼠肉瘤生长的抑制作用。采用ELISA法测定细胞因子IL-6和TNF-α的水平,HE染色观察肉瘤病理切片,探讨EALs对荷瘤小鼠免疫功能的影响。结果EALs对人乳腺癌细胞MCF-7、人肝癌细胞Hep G2、人慢性髓原白血病细胞K562、小鼠腹水瘤细胞S180、人非小细胞肺癌细胞A549、人组织细胞淋巴瘤细胞U937这6种常见的人肿瘤细胞均有抑制作用,其IC50值在95.123~303.427μg·mL-1。EALs对人乳腺癌细胞MCF-7抑制效果最好;EALs可明显抑制荷S180肉瘤小鼠模型肉瘤生长,能降低荷瘤小鼠血清细胞因子IL-6和TNF-α的水平,病理切片显示,中浓度EALs抑瘤效果较好。结论EALs具有一定的体内外抗肿瘤活性,体外人乳腺癌细胞MCF-7对其最为敏感,体内抑制效果以中浓度最佳,其作用机制可能与促进免疫功能有关。Objective To determine the antitumor activity of water extract of Elaeagnus angustifolia L.leaves(EALs)from Xinjiang in vitro and in vivo.Methods The antitumor ectivity of EALs on tumor cells was evaluated by MTT in vitro and IC50 was calculated.The inhibition of EALs on tumor growth in vivo was observed by the models of implanted sarcoma 180(S180)in mice.The levels of IL-6 and TNF-αwere measured by ELISA,and the pathological sections of sarcomas were observed by HE staining to determine the immune function of EALs on tumor bearing mice.Results EALs had inhibition effect on MCF-7,HepG2,K562,S180,A549 and U937 cells in vitro,and the IC50 values ranged from 95.123 to 303.427μg·mL-1.The inhibition effect of MCF-7 cells was the best.The inhibition of EALs on the growth of S180 mice were significant in vivo.EALs reduced the levels of IL-6 and TNF-αin the serum of tumor bearing mice.The pathological section showed that EALs of middle concentration had better antitumor effect.Conclusion EALs have a certain antitumor activity in vitro and in vivo,and the most sensitive tumor cell is MCF-7 cells.Medium concentration of EALs shows the best inhibition in vivo,which is related to the promotion of immune function.
