摘　　要：目的建立HPLC-ELSD法同时测定铁笛丸中甘草酸、桔梗皂苷D、贝母素甲与贝母素乙的含量。方法采用高效液相色谱法,色谱柱为Venusil MP C18,流动相为甲醇-水(梯度洗脱),流速为1.0 mL·min-1,检测器为蒸发光散射检测器,柱温为30℃,进样量为10μL。结果甘草酸、桔梗皂苷D、贝母素甲与贝母素乙检测线性范围分别为12.305~1230.5μg·mL^-1(r=0.999)、2.019~201.9μg·mL^-1(r=0.999)、2.004~200.4μg·mL^-1(r=0.999)、1.294~129.4μg·mL^-1(r=0.999);定量限分别为0.62、0.50、0.50、0.65μg·mL^-1;检测限分别为0.20、0.17、0.17、0.22μg·mL^-1;平均加样回收率分别为93.8%(RSD=1.3%)、96.5%(RSD=1.6%)、97.2%(RSD=1.2%)、94.3%(RSD=1.2%)。结论本方法简便、准确,适用于铁笛丸中甘草酸、桔梗皂苷D、贝母素甲与贝母素乙含量的同时测定。Objective To simultaneously determine glycyrrhizic acid,platycodin D,peimine,and peiminine in Tiedi pills by HPLC-ELSD.Methods HPLC method was adopted.The separation was performed on a Venusil MP C18 column with mobile phase consisting of methanol-water(gradient elution)at the flow rate of 1.0 mL·min-1.The detector was evaporative light scattering detector.The column temperature was 30℃.The injection volume was 10μL.Results The linear ranges were 12.305~1230.5μg·mL^-1 for glycyrrhizic acid(r=0.999),2.019~201.9μg·mL^-1 for platycodin D(r=0.999),2.004~200.4μg·mL^-1 for peimine(r=0.999),and 1.294~129.4μg·mL^-1 for peiminine(r=0.999),respectively.The limits of quantitation were 0.62,0.50,0.50,and 0.65μg·mL^-1,and the limits of detection were 0.20,0.17,0.17,and 0.22μg·mL^-1,respectively.The average recoveries were 93.8%(RSD=1.3%),96.5%(RSD=1.6%),97.2%(RSD=1.2%),and 94.3%(RSD=1.2%),respectively.Conclusion The method is simple and accurate,and can be used for the simultaneous determination of glycyrrhizic acid,platycodin D,peimine,and peiminine in Tiedi pills.
