摘  要:[目的]采用聚氨酯构建慢性颈脊髓压迫动物模型,观察受压颈脊髓内早期炎症反应。[方法]SD大鼠随机均分为颈髓压迫模型(CSM)组和对照组。(CSM)组C6椎板下置入新型吸水性可膨胀生物材料聚氨酯,对照组不置入可膨胀性生物材料。建模14 d后行颈椎核磁共振检查,观察大鼠脊髓受压影像学表现。建模3、7、14 d后分别取大鼠受压颈脊髓组织,采用蛋白印迹方法检测受压颈脊髓组织p-MK2、炎症因子(NF-κB)、凋亡相关蛋白(caspase-8、Bax/Bcl-2)的变化。[结果]大鼠颈椎MRI检查提示CSM组矢状位、轴位MRI可见脊髓明显受压、局部变细。CSM组SD大鼠p-MK2、NF-κB、caspase-8和Bax表达较对照组显著增多(P<0.05),而Bcl-2的表达则显著降低(P<0.05)。[结论]聚氨酯构建的动物模型中受压的颈脊髓内早期出现了炎症反应,MK2可能参与了炎症反应。[Objective]To establish an animal model of chronic cervical spinal cord compression with polyurethane,and then observe the early inflammatory reaction of cervical spinal cord tissue under chronic compression.[Methods]SD rats were randomly divided into chronic spinal compression model(CSM)group and control group.The polyurethane,a new water-absorbing expansion material,was implanted under C6/7 lamina to prepare chronic cervical spinal cord compression in the CSM group,whereas nothing was implanted in the control group.At 14 days after operation,MRI examinations were conducted on the animals to reveal the imaging presentation.After the rats were sacrificed at 3,7 and 14 days after operation,the homogenate of involved spinal cord segment was prepared to assay signal path MK2 protein,inflammatory factor(NF-κB),apoptosis-related protein(Caspase-8,Bcl-2/Bax)by Western blotting.[Results]The cervical MRI examination showed that spinal cord was pressured by the expended polyurethane on the sagittal and transverse planes.The Western blotting assay revealed that the CSM group had significant increases of MK2,NF-κB,Caspase-8 and Bax(P<0.05),whereas significant decrease of Bcl-2 compared with those of the control group(P<0.05).[Conclusion]The early inflammatory response is involved in the chronic cervical spine compression model created by polyurethane,which is related to the MK2 signaling pathway.
