油茶果实发育相关基因SPATULA/ALCATRAZ的鉴别

摘　　要：拟南芥(Arabidopsis thaliana)SPATULA/ALCATRAZ是与果实发育和果实大小有关的基因。对普通油茶(Camellia oleifera)幼叶、未成熟种仁、花蕾的转录组数据进行挖掘,得到2条油茶SPATULA基因序列和1条油茶ALCATRAZ基因序列;从油茶基因组三代测序数据中鉴别出含有前述3条序列的基因组DNA序列;最后,得到3条油茶SPATULA/ALCATRAZ基因全长CDS序列,分别命名为CoSPT1、CoSPT2、CoALC。用基部被子植物无油樟(Amborella trichopoda)的SPATULA及核心真双子叶植物53个SPATULA/ALCATRAZ蛋白质序列构建进化树,结果表明,CoALC与拟南芥AtALC分布在不同的亚支,CoALC在油茶中的功能可能与AtALC不同,油茶蒴果的开裂机制可能与拟南芥不同;CoSPT1和CoSPT2的序列相似度只有54.57%,并且与CacuaSPT在同一个亚支,CoSPT1和CoSPT2可能是72.9~101.4百万年前Ad-β全基因组复制的产物。SPATULA/ALCATRAZ genes are related to fruit development and fruit size in Arabidopsis thaliana.Two CDSs homologous to SPATULA and a CDS homologous to ALCATRAZ were obtained by mining transcriptome data from Camellia oleifera young leaf,immature kernels and flower buds.Then,3 long DNA fragments containing the CDS were identified from the whole genome SMRT sequence data.Finally,3 full-length CDSs were identified and named CoSPT1,CoSPT2,CoALC,respectively.A SPATULA protein from basal angiosperm Amborella trichopoda and 53 SPATULA/ALCATRAZ proteins from the core eudicots were employed to construct a phylogenetic tree,which revealed that CoALC and AtALC were distributed in different clans which suggested that CoALC might have different functions compared with AtALC and there were existed different mechanisms during the fruit dehiscence in C.oleifera and A.thaliana.CoSPT1 and CoSPT2 might be products of Ad-βwhole genome duplication event occurred 72.9-101.4 million years ago because the similarity between CoSPT1 and CoSPT2 was 54.57%,and the phylogenetic analysis suggested that CoSPT1,CoSPT2 and CacuaSPT were in a clan.
