摘 要:目的探讨子痫前期孕妇胎盘组织中微小RNA-34a(microRNA-34a,miR-34a)表达改变的临床意义。方法选取东莞市人民医院2017年9月至2018年6月诊治的子痫前期产妇30例,另取正常行剖宫产的产妇30例作为对照。实时荧光定量PCR检测miR-34a和髓细胞白血病因子-1(myeloid cell leukemia-1,MCL-1)的表达,蛋白质印迹(Western blot)检测MCL-1蛋白和自噬相关蛋白微管相关蛋白I轻链3Ⅱ(microtubule associated protein 1 lightchain 3Ⅱ,LC3Ⅱ)的表达。计量资料采用t检验,相关性比较采用Spearman相关分析。结果子痫前期组胎盘组织中,其miR-34a的2-△△Ct值为0.217±0.035,高于对照组的0.126±0.028(P<0.01);MCL-1的2-△△Ct值为0.208±0.037,低于对照组的0.316±0.054(P<0.01);miR-34a表达与MCL-1表达呈负相关(r=-0.412,P<0.05);MCL-1蛋白表达灰度值为0.413±0.072,低于对照组的0.826±0.142(P<0.01);LC3Ⅱ蛋白表达灰度值为0.316±0.048,高于对照组的0.152±0.024(P<0.01)。Spearman相关分析显示子痫前期组胎盘组织中LC3Ⅱ蛋白表达与MCL-1蛋白表达呈负相关(r=-0.607,P<0.05),与miR-34a表达呈正相关(r=0.582,P<0.05)。结论miR-34a在子痫前期患者胎盘组织中高表达,可能抑制MCL-1基因,激活自噬,参与子痫前期的致病过程。Objective To explore the clinical significance of microRNA-34a(miR-34a)expression in placenta of the patients with preeclampsia.Methods Thirty patients with preeclampsia in Dongguan People's Hospital between September 2017 and June 2018 were enrolled in this study.Another 30 cases of normal cesarean section pregnant women were selected as control group.Real-time PCR was used to detect miR-34a and myeloid cell leukemia-1(MCL-1).MCL-1 protein and microtubule associated protein 1 lightchain 3Ⅱ(LC3Ⅱ)were detected by Western blot.Quantitative data were compared with t test,and the comparative study was carried by Spearman correlation analysis.Results In preeclampsia group,the 2-△△Ct value of miR-34a was 0.217±0.035,which was significantly higher than that of 0.126±0.028 in control group(P<0.01).The 2-△△Ct value of MCL-1 in preeclampsia group was 0.208±0.037,which was significantly lower than that of 0.316±0.054 in control group(P<0.01).The expression of miR-34a in preeclampsia group was negatively correlated with MCL-1 expression(r=-0.412,P<0.05).The gray level of MCL-1 protein in preeclampsia group was 0.413±0.072,which was significantly lower than that of 0.826±0.142 in control group(P<0.01).The gray level of LC3Ⅱprotein in preeclampsia group was 0.316±0.048,which was significantly higher than that of 0.152±0.024 in control group(P<0.01).Spearman correlation analysis showed that there was a significant negative correlation between LC3Ⅱprotein expression and MCL-1 protein expression(r=-0.607,P<0.05),and LC3Ⅱhad a significant positive correlation with miR-34a expression(r=0.582,P<0.05).Conclusion High expression of miR-34a in placenta of preeclampsia may inhibit MCL-1 gene to promote autophagy,and participate in the pathogenesis of preeclampsia.
