摘  要:钙网蛋白(Calreticulin,CRT)是一种内质网膜或肌质网膜上的钙离子结合蛋白,主要参与调控蛋白质的正确折叠,维持昆虫体内钙离子的平衡以及昆虫免疫反应.基于稻纵卷叶螟(Cnaphalocrocis medinalis)的转录组数据库,我们鉴定到两个Calreticulin基因,分别命名为CmCRT1和CmCRT2.CmCRT1开放阅读框为3194 bp,编码397个氨基酸;CmCRT2的开放阅读框为1206 bp,编码401个氨基酸.进化树分析显示CmCRT1主要与大蜡螟CRT有较高的同源性,而CmCRT2主要与丽蝇蛹集金小峰的CRT保持较高的亲缘关系.组织表达分析显示CmCRT在血淋巴中具有较高的表达水平,其次是脂肪体及中肠.发育时期表达分析表明CmCRTs主要在稻纵卷叶螟的蛹期及成虫期表达.此外,CmCRT基因经大肠杆菌(Escherichia coli)刺激后可被显著诱导表达.综上所述,本研究为进一步研究稻纵卷叶螟CRT的功能奠定基础.Calreticulin is a Ca^2+ binding protein of endoplasmic reticulum or sarcoplasmic reticulum that mainly involved in directing proper conformation of proteins,regulating Ca^2+ homoeostasis and involving in immune response.In this study,we identified two calreticulin(CmCRT1 and CmCRT2)gene from transcriptome data,which encoded 397 and 401 amino acids,respectively.Phylogenetic analysis indicated that the CmCRT1 had the close genetic distance with GmCRT(Lepidoptera),while CmCRT2 had close relationship with PpCRT and NvCRT(Hymenoptera).Tissue expression analysis indicated that CmCRT had a significantly higher expression in the haemolymph,followed by fat body and midgut.The developmental stage expression analysis showed that CmCRT was mainly expressed in pupa and adult.CmCRT mRNA expression and protein expression level were significantly induced by Escherichia coli.These data indicated that CmCRTs may play an important role in innate immune responses of C.medinalis.
