摘 要:目的:利用中药系统鉴定法鉴别青葙子及其混伪品,为青葙子药材的准确鉴定及质量控制提供参考。方法:通过性状、微性状、显微鉴别和DNA条形码分析等方法,对青葙子药材及其混伪品进行比对分析,利用MEGA 7.0软件计算物种DNA条形码序列的种内、种间Kimura 2-Parameter遗传距离、评估序列的条形码间距,采用邻接法构建系统聚类树。结果:仅凭性状鉴别难以区分青葙子与混伪品;青葙子及其混伪品的微性状、显微特征差异明显,可作为其鉴定的重要依据之一;经筛选,推荐ITS序列作为青葙子及其混伪品的DNA条形码候选序列,trnL序列作为补充序列。结论:中药系统鉴定法能够完成对青葙子药材的精准鉴别。Objective:To identify Celosia argentea and its adulterants by systematic identification of traditional Chinese medicine,in order to provide reference for the accurate identification and quality control of Celosia argentea.Methods:Through the methods of characterization,microcharacterization,microscopic identification and DNA barcoding analysis,the comparative analysis of Celosia argentea and its adulterants was carried out.MEGA 7.0 software was used to calculate the genetic distance of in-species and inter-species Kimura 2-Parameter of DNA barcoding sequences of species and evaluate the barcoding gap of sequences,the system cluster tree was constructed by neighbor-joining method.Results:It was difficult to distinguish Celosia argentea and its adulterants only by character identification.The differences in microcharacters and microscopic characteristics of Celosia argentea and its adulterants were obvious,which could be used as an important basis for identification.After screening,the ITS sequence was recommended as the candidate sequence of DNA barcoding of Celosia argentea and its adulterants,and the trnL sequence was used as the supplementary sequence.Conculsion:The systematic identification of traditional Chinese medicine can be used for accurate identification of Celosia argentea.
