摘 要:目的探讨组织型转谷氨酰胺酶(tTG)在大鼠心肌梗死后心肌纤维中的表达,分析其在心肌梗死后心肌纤维化中的作用与意义。方法选取30只8周龄雄性SD大鼠,随机均分为心肌梗死组和假手术组2组,其中心肌梗死组采用冠状动脉(冠脉)左前降支结扎方法构建,假手术组只缝针不结扎冠脉。造模4周后超声心动图检测心功能,检测tTG与纤维蛋白1在RNA水平中的表达,tTG及纤维蛋白1在蛋白水平的表达。结果与假手术组相比,心肌梗死组大鼠心室明显扩张[左心室收缩末期内径(LVESD):(4.88±0.36)mm vs.(1.79±0.28)mm,左心室舒张末期内径(LVEDD)为(6.36±0.42)mm vs.(3.25±0.32)mm],差异均有统计学意义(P<0.05);心功能严重受损[左心室射血分数(EF):(80.75±5.76)%vs.(28.64±3.14)%,短轴缩短率(FS):(42.23±4.13)%vs.(21.51±2.32)%],差异均有统计学意义(P<0.05);心肌组织纤维化程度加重,心肌胶原容积分数增加[(2.4±0.25)%vs.(11.7±0.32)%],存在统计学差异(P<0.05);与正常大鼠相比,患有心肌梗死的大鼠心肌组织中tTG的转录水平和蛋白水平显著增加,同时纤维蛋白1的表达在转录水平和蛋白水平显著增加,存在统计学差异(P<0.05)。结论转谷氨酰胺酶在梗死后心肌纤维化过程中发挥了重要作用,其与纤维蛋白1共同参与心肌纤维化的过程;转谷氨酰胺酶可能成为心肌纤维化诊断和治疗靶点。Objective To investigate the expression of tissue-type transglutaminase(tTG)in myocardial fibers in rats after myocardial infarction(MI),and analyze the effect and significance of tTG in myocardial fibrosis.Methods SD rats(n=30,aged 8 weeks old)were selected and randomly divided into MI group and sham-operation group.The MI group was given ligation of coronary left anterior descending branch for modeling,and shamoperation group was just given stitch without ligation.After modeling for 4 weeks,heart function was examined by using echocardiogram,and the mRNA and protein expressions of tTG and fibrin 1 were detected.Results Compared with sham-operation group,there was significant ventricular dilation[LVESD:(4.88±0.36)mm vs.(1.79±0.28)mm,LVEDD:(6.36±0.42)mm vs.(3.25±0.32)mm]in MI group(P<0.05),severe damage of heart function[LVEF:(80.75±5.76)%vs.(28.64±3.14)%,LVFS:(42.23±4.13)%vs.(21.51±2.32)%in MI group(P<0.05)],and exacerbation of myocardial fibrosis and increase of myocardial collagen volume fraction[(2.4±0.25)%vs.(11.7±0.32)%]in MI group(P<0.05).The mRNA and protein expressions of tTG and fibrin 1 increased significantly in MI group compared with sham-operation group(P<0.05).Conclusion The effect of tTG is important in the process of myocardial fibrosis after MI,and tTG and fibrin 1 take part in myocardial fibrosis together.Tissue-type transglutaminase may be a diagnostic and therapeutic target of myocardial fibrosis.
