桃金娘鞣质抗腹泻作用机理研究

摘　　要：研究桃金娘鞣质抗腹泻作用机理。以正常小鼠为模型,测试胃内容物排空率、小肠内容物推进率,观察桃金娘鞣质对胃肠推进运动的影响;以乙酸注射腹腔炎症小鼠为模型,测试血液中伊文斯蓝腹腔透入量,观察桃金娘鞣质对腹腔毛细血管通透性影响;以豚鼠离体肠道为模型,测试生理收缩反应峰值、乙酰胆碱诱导收缩反应峰值,观察桃金娘鞣质对肠道平滑肌收缩反应的影响。结果显示,桃金娘鞣质能显著抑制小鼠的胃排空运动及小肠推进运动(P<0.05);显著降低腹腔炎症小鼠血液中伊文斯蓝腹腔透入量(P<0.05);显著抑制豚鼠离体肠道生理收缩反应(P<0.05),不能拮抗乙酰胆碱诱导的收缩反应(P>0.05)。说明桃金娘鞣质主要通过抑制胃肠收缩运动或降低肠道蠕动;降低腹腔毛细血管、肠绒毛膜通透性;抑制炎症介质合成释放或抑制炎症介质泻下作用等机制产生抗腹泻作用;与阿片激动剂类药物配伍使用抗腹泻效果可能更好。Study the anti-diarrhea mechanism on the base myrtle tannin.The effects of tannin from Myrtle on gastrointestinal motility were observed by measuring the gastric emptying rate and the intestinal pushing rate of normal mice.In order to observe the effect of tannin from Myrtle on the permeability of capillaries in the abdominal cavity,acetic acid injected intraperitoneal inflammatory mice as the model were used to test the amount of Evans blue intraperitoneal penetration in the blood.The effects of tannin from Myrtle on contractile response of intestinal smooth muscle the peak of physiological contractile response and the peak of acetylcholine induced contractile response were measured in isolated small intestine of guinea pig.The results showed that the tannin from Myrtle could significantly inhibit gastric emptying and small intestine propulsion in mice(P<0.05),significantly reduce the amount of Evans blue intraperitoneal penetration in the blood of mice with abdominal inflammation(P<0.05),and significantly inhibit the physiological contractile response of isolated intestine in guinea pigs(P<0.05),but can not antagonize the contraction induced by acetylcholine(P>0.05).The antidiarrheal effect of tannin from Myrtle is mainly through inhibiting gastrointestinal contractile movement or intestinal peristalsis,reducing the permeability of capillaries and chorionic villi in abdominal cavity,anti diarrhea effect is produced by inhibiting the synthesis and release of inflammatory mediators or the diarrhea of inflammatory mediators,and the antidiarrheal effect may be better if combined with opioid agonists.
