康复新液对乳腺腺病患者乳腺腺体组织脂筏结构蛋白1的影响

摘　　要：目的观察脂筏结构蛋白1(FLOT1)在乳腺腺病患者乳腺腺体组织中的表达,以及康复新液对其表达的影响。方法采用干预观察性研究方法,选取病理科标本库中96例正常乳腺标本作为空白对照组,收集乳腺腺病患者96例为乳腺腺病组,经康复新液口服治疗1年,每次10ml,每天3次。治疗前后采用放射免疫沉淀法、免疫印迹法检测乳腺腺病组患者及空白对照组乳腺腺体组织中FLOT1蛋白表达;比较乳腺腺病组患者治疗前后FLOT1基因检测阳性率,并行治疗前后乳腺彩超评分。结果乳腺腺病组患者治疗1年后乳腺彩超评分明显低于治疗前(P<0.05)。乳腺腺病组患者治疗前乳腺腺体组织中FLOT1蛋白表达量、基因强度评分均高于空白对照组(P<0.05或P<0.01),乳腺腺病组患者治疗后FLOT1蛋白表达量与空白对照组差异无统计学意义(P>0.05)。乳腺腺病组患者治疗后FLOT1基因检测阳性率为43.75%,明显低于治疗前的97.92%(P<0.01)。结论口服康复新液可以改善乳腺腺病患者乳腺彩超评分,并且抑制腺体中FLOT1的高表达。Objective To observe the expression of flotillin-1(FLOT1)in the breast glandular tissues in patients with adenosis of breast,and the effects of Kangfuxin Liquid on its expression.Methods In the inventive,observative study,96 normal breast specimens collected from tissue bank of the department of pathology were set as blank control group,and 96 patients with adenosis of breast were set as adenosis group,in which Kangfuxin Liquid was orally administered,10 ml each time,three times a day,for one year.Radioimmuno precipitation and western blotting were used to detect the expression of FLOT1 in the breast glandular tissues of patients in adenosis group and that of breast specimens in the blank control group before and after treatment.FLOT1 gene detection positive rate,as well as the breast color doppler ultrasound score before and after treatment in the adenosis group before treatment were compared.Results The breast color doppler ultrasound score was significantly lower after one-year treatment compared to that before treatment(P<0.05).The expression of FLOT1 protein and gene intensity score in adenosis group before treatment were higher than those in the blank control group(P<0.05 or P<0.01).There was no statistical differences between two groups in expression of FLOT1 protein after treatment(P>0.05).The positive rate of FLOT1 gene detection in adenosis group was 43.75%after treatment,significantly lower than 97.92%before treatment(P<0.01).Conclusion Oral administration of Kangfuxin Liquid could increase color doppler ultrasound score and inhibit the over-expression of FLOT1 in the glandular tissue.
