摘 要:目的探讨安神定志灵方对注意力缺陷多动障碍(ADHD)的作用机制。方法将自发性高血压大鼠(SHR)40只随机分为模型组、择思达组和安神定志灵低、中、高剂量组,每组8只,另设Wistar京都大鼠(WKY)8只为正常1组,SD大鼠8只为正常2组。择思达组给予盐酸托莫西汀胶囊0.045mg/(kg·d)灌胃,安神定志灵低、中、高剂量组分别给予安神定志灵方6.7、13.4、26.7g/(kg·d)灌胃,正常1组、正常2组、模型组均灌胃生理盐水1.5ml/100g,均每日早晚2次灌胃,持续4周。运用蛋白免疫印迹法检测大鼠前额叶皮质多巴胺-β-羟化酶(DβH)、去甲肾上腺素转运体(NET)蛋白表达,运用PCR技术检测DβH、NET mRNA表达,运用免疫荧光技术检测DβH、NET阳性细胞表达。结果与正常1组、正常2组比较,模型组大鼠前额叶皮质DβH蛋白、mRNA表达及阳性细胞表达量降低,NET蛋白、mRNA表达及阳性细胞表达量升高(P<0.05);与模型组比较,择思达组及安神定志灵低剂量组DβH蛋白、mRNA表达及阳性细胞表达量升高,择思达组和安神定志灵低、中剂量组NET蛋白、mRNA表达及阳性细胞表达量降低(P<0.05);与择思达组比较,安神定志灵低剂量组DβH蛋白、mRNA表达及阳性细胞表达量升高,安神定志灵高剂量组NET蛋白、mRNA表达及阳性细胞表达量降低(P<0.05)。结论安神定志灵方可能通过调控SHR大鼠前额叶皮质DβH、NET的表达,达到控制注意力缺陷多动障碍临床症状的目的。Objective To investigate the mechanism of Anshen Dingzhi Ling Formula(安神定志灵方)on attention deficit hyperactivity disorder(ADHD).Methods Totally 40 spontaneously hypertension rats(SHR)were randomly divided into a model group,a strattera group,Anshen Dingzhi Ling low,medium and high dose groups,with eight rats in each group.Additionally,eight Wistar-Kyoto rats(WKY)were set as normal group 1,and eight SD rats were set as normal group 2.The rats in strattera group were administered with atomoxetine hydrochloride capsule 0.045 mg/kg per day;Anshen Dingzhi Ling low,medium and high dose groups were administered with 6.7,13.4,and 26.7 g/kg Anshen Dingzhi Ling per day separately;and rats in the normal group 1,normal group 2 and model group were given normal saline 1.5 ml/100 g;all groups were administered by gavage,twice daily,every morning and evening,for 4 weeks.The expression of dopamineβ-hydroxylase(DβH)and norepinephrine transporter(NET)protein in the prefrontal cortex were detected by Western blotting;the expression of DβH and NET mRNA was detected by PCR technology;and the expression of DβH and NET positive cells were detected by immunofluorescence technology.Results Compared with normal group 1 and normal group 2,the expression of DβH protein,mRNA,and positive cells in the prefrontal cortex of model group were decreased,while that of NET protein,mRNA,and positive cells were increased(P<0.05).Compared with the model group,the expression levels of DβH protein,mRNA,and positive cells were higher in strattera group and in Anshen Dingzhi Ling low dose group,and the expression levels of NET protein,mRNA,and positive cells were lower in strattera group and in Anshen Dingzhi Ling low and medium dose groups(P<0.05).Compared with strattera group,the expression level of DβH protein,mRNA,and positive cells were higher in Anshen Dingzhi Ling low dose group,while the expression level of NET protein,mRNA,and positive cells were lower in Anshen Dingzhi Ling high dose group(P<0.05).Conclusion Anshen Dingzhi Ling
