摘 要:目的:研究低剂量亚慢性苯染毒对小鼠的毒性作用,探讨橙皮精油对中毒小鼠的保护作用及作用机制。方法:取30只清洁级健康ICR小鼠,随机分为对照组、苯染毒组和苯+精油组(10只/组)。其中,苯染毒组以200 mg·kg^(-1)体重的剂量行苯灌胃染毒,苯+精油组给予苯+精油的混合溶液,对照组给予等剂量的乙醇溶液。均染毒30 d。比较三组的体重变化情况、各脏器(心、肝、肾、脾、睾丸、卵巢、子宫等)的脏器系数、干预前后的血常规情况[包括红细胞计数(RBC)、白细胞计数(WBC)、血红蛋白(HGB)和血小板(PLT)]及各组血清和肝组织中的抗氧化指标含量[包括超氧化物歧化酶(SOD)、丙二醛(MDA)]。结果:对照组小鼠体重呈持续缓慢增长,苯染毒组体重下降明显,苯+精油组体重较为平稳、略有下降;三组小鼠肝脏、脾脏和肺的脏器系数均表现为:苯染毒组>苯+精油组>对照组(P<0.05),而心脏、肾脏的脏器系数相比差异无统计学意义(P>0.05);在各血常规指标(RBC、HGB和PLT)及血、肝SOD指标方面,苯染毒组<苯+精油组<对照组(P<0.05);而在血和肝MDA水平上,苯染毒组>苯+精油组>对照组(P<0.05)。结论:低剂量亚慢性苯染毒可对小鼠的血液系统和抗氧化指标造成损伤作用,橙皮精油具有有效的改善作用。Objective:To study the toxic effect of low dose subchronic benzene on mice,and to explore the protective effect and mechanism of orange peel essential oil on poisoned mice.Methods:Thirty clean healthy ICR mice were selected and randomly divided into control group,benzene poisoning group and benzene+essential oil group according to random block method(10 mice in each group).Among them,the benzene poisoning group was poisoned by intragastric administration of benzene at a dose of 200 mg·kg-1 body weight,the benzene+essential oil group was given a mixed solution of benzene+essential oil,and the control group was given an equal dose of ethanol solution.All were infected for 30d.The situation of weight change,viscera coefficient of each organs(heart,liver,kidney,spleen,testis,ovary and uterus,etc.),blood routine before and after the intervention[including red blood cell count(RBC),white blood cell count(WBC),hemoglobin(HGB)and platelet(PLT)]and antioxidant index content in serum and liver tissue among groups[including superoxide dismutase(SOD),malondialdehyde(MDA)]were compared among the three groups.Re⁃sults:The body weight of mice in the control group showed a sustained and slow growth,and the body weight of the benzene poisoning group decreased significantly,and the body weight of the benzene+essential oil group was stable and slightly decreased.The organ coefficients of liver,spleen and lung of the three groups of mice were shown as follows:benzene poisoning group>benzene+essential oil group>control group(P<0.05),while the organ coefficients of heart and kidney showed no statistical difference(P>0.05).In terms of blood routine indexes(RBC、HGB and PLT)and SOD index in blood and liver,the order of contents was showed as follows:The benzene poisoning group<benzene+essential oil group<control group(P<0.05);While on the blood and liver MDA levels,the order of contents was showed as follows:the benzene poisoning group>benzene+essential oil group>control group(P<0.05).Conclu⁃sion:Low dose of subchronic benzene cou
